摘  要:目的观察5-HT6R激动剂对血管性痴呆大鼠认知功能的影响。方法健康雄性SD大鼠采用双侧颈总动脉永久性结扎法建立血管性痴呆(VD)模型。给予5-HT6R激动剂治疗后,观察VD大鼠海马组织的病理变化,Morris水迷宫测试VD大鼠的认知功能,测定大鼠血清中MDA含量、GSH-Px活性。结果与假手术组比较,模型组大鼠海马区细胞排列紊乱,细胞核固缩,核仁不清晰,逃避潜伏期延长,穿台次数减少(P<0.05),血清中MDA含量增加,GSH-Px活性降低(P<0.05)。与模型组比较,5-HT6R激动剂组大鼠的海马区脑组织病理结构得到改善,认知功能有所提升,血清中MDA含量降低,GSH-Px活性升高(P<0.05)。结论 5-HT6R激动剂可改善血管性痴呆大鼠的认知功能。Objective To explore the effects of 5-hydroxytryptamine 6 receptor agonist on cognitive function in vascular dementia rats.Methods The model of vascular dementia was established by permanent occlusion of bilateral carotid arteries in healthy male Sprague-Dawley rats.After treatment with 5-HT6 R agonist,pathological changes in hippocampal tissue of VD rats were observed.The cognitive functions of VD rats were tested by Morris water maze,and the content of MDA and the activity of GSH-Px in the serum of rats were determined.Results Compared with the sham-operated group,the model group had disordered cell arrangement in the hippocampus,nucleus shrinkage,unclear nucleolus,prolonged escape latency,reduced number of penetrations(P<0.05),increased serum MDA content,and decreased GSH-Px activity(P<0.05).Compared with the model group,the pathological structure of hippocampus tissue of the rats in the 5-HT6 R agonist was improved,and their cognitive function was improved,the MDA content in serum was decreased,and the GSH-Px activity was increased(P<0.05).Conclusion 5-HT6 receptor agonist can improve the cognitive function of rats with vascular dementia.
