联合检测RCAS1和CA19-9对胰腺癌的诊断价值

摘　　要：[目的]检测胰腺癌患者血清和组织中RCAS1和CA19-9的表达,以探讨RCAS1和CA19-9联合检测在胰腺癌诊断中的价值。[方法]采用ELISA法分别检测43例胰腺癌患者(胰腺癌组)、30例正常人(正常对照组)血清中RCAS1和CA19-9含量。采用Western Blot法和RT-PCR法检测20对胰腺癌组织和癌旁组织中RCAS1和CA19-9的表达。综合评价联合检测RCAS1和CA19-9对诊断胰腺癌的价值。[结果]ELISA法检测胰腺癌组TNM分期I+Ⅱ期的患者血清RCAS1和CA19-9明显高于正常对照组(P<0.01)。Ⅲ+Ⅳ期的患者血清RCAS1和CA19-9明显高于I+Ⅱ期(P<0.05)。RCAS1和CA19-9联合检测胰腺癌的灵敏度是95.3%,较单独检测CA19-9明显增高(P<0.05)。Western Blot和RT-PCR结果示胰腺癌组织中RCAS1和CA19-9的表达均明显高于癌旁组织(P<0.05)。[结论]胰腺癌患者血清和组织中RCAS1和CA19-9表达明显增加,两者联合检测可提高胰腺癌的早期诊断率,对诊断胰腺癌及判断预后有重要价值。[Objective]To detect the expression of RCAS1 and CA19-9 in serum and tissues of patients with pancreatic cancer and evaluate the value of combined detection of RCAS1 and CA19-9 in diagnosis of pancreatic cancer.[Method]The serum content of RCAS1 and CA19-9 in 43 patients with pancreatic cancer and 30 normal individuals were detected by ELISA.The expression of RCAS1 and CA19-9 in 20 pairs of pancreatic cancer tissues and cancer-adjacent tissues were detected by Western Blot and RT-PCR.The application of both biomarkers in diagnosis of pancreatic cancer was evaluated.[Results]The serum RCAS1 and CA19-9 in pancreatic cancer group with stage I+Ⅱof TNM staging were significantly higher than that in normal group by ELISA(P<0.01).The serum RCAS1 and CA19-9 in pancreatic cancer group with stageⅢ+Ⅳwere significantly higher than those in stage I+Ⅱ(P<0.05).The sensitivity of the combined detection of RCAS1 and CA19-9 was 95.3%,which was significantly higher than that of CA19-9 alone(P<0.05).Western Blot and RT-PCR results demonstrated that the expression of RCAS1 and CA19-9 in pancreatic cancer tissues were significantly higher than those in cancer-adjacent tissues(P<0.05).[Conclusion]Obvious increase of RCAS1 and CA19-9 levels was observed in serum and tissues of patients with pancreatic cancer.The detection of both biomarkers can improve the early diagnostic rate of pancreatic cancer,which provides significant value in the diagnosis and prognosis of pancreatic cancer.
