摘  要:目的:构建并评估肝纤维化骨质疏松大鼠模型。方法:2016年9月至2018年9月,将40只雄性SD大鼠(南京医科大学实验动物中心提供),采用随机数字表法分为假手术4周(4W)、假手术6W、胆总管结扎(BDL)4W、BDL6W组。实验动物分别行假手术及胆总管结扎术后4周及6周后检测血清肝纤维化指标,获取肝脏及股骨标本并行肝脏组织马松(Masson)染色分析肝纤维化、股骨微计算机断层扫描技术(Micro CT)分析骨质疏松。组间比较采用单因素方差分析结合Bonferroni’s多重比较检验。结果:血清肝纤维化指标检测结果显示除血清透明质酸酶外[(1368.48±1093.82)U/L比(108.785±17.69)U/L,t=3.277,P<0.05],谷丙转氨酶(AST)、谷草转氨酶(ALT)、血清Ⅳ型胶原蛋白在BDL4W和假手术4W之间差异均无统计学意义[(317.73±98.69)U/L比(212.13±58.51)U/L,(81.03±21.82)U/L比(41.1±21.19)U/L,(19.81±0.42)μg/L比(19.35±0.34)μg/L,t=1.586、1.007、0.938,P值均>0.05);而BDL6W组各血清学指标均显著高于假手术6W组(t=5.845、3.442、4.928、3.754,P值均<0.05)。肝脏Masson染色结果显示BDL4W大鼠出现肝纤维化改变,BDL6W大鼠更为明显。与假手术组大鼠比较,BDL6W大鼠Micro CT结果显示具有明显骨质疏松改变。结论:大鼠胆总管结扎术可构建肝脏纤维化及相应骨质疏松动物模型。Objective The purpose of this study was to construct and evaluate a rat model of primary biliary cirrhosis related osteoporosis,which could be used for the study of the pathogenesis and treatment strategies ofhepatic cirrhosisrelated bone loss.Methods The research was carried out in Nanjing Drum Tower hospital between September,2016 and September,2018.Forty male SD rats(Nanjing Medical University,Qualification No.201901030,weight 200-250 g)were randomly divided into four group susing the random number table method:sham operation 4 weeks(SO4W),SO6W,bile duct ligation(BDL)4W,and BDL6W groups.Serum liver fibrosis indexes were examined in experimental animals 4 and 6 weeks after surgery.The liver and femur specimens were obtained after the animals were sacrificed.Masson staining was performed to evaluate liver cirrhosis.Micro-computed temography(Micro CT)was used to assess bone loss in experimental animals.The results are presented as the means and standard deviations(SDs)and were analyzed by one-way ANOVA with Bonferroni’s multiple comparison test.Results There was no significantly statistical difference towards serum liver fibrosis index between animals in BDL4W and SO4W groups in terms of aspartate aminotransferase(AST),alanine aminotransferase(ALT)and Serum typeⅣcollagen(317.73±98.69)U/L vs.(212.13±58.51)U/L,(81.03±21.82)U/L vs.(41.1±21.19)U/L,(19.81±0.42)μg/L vs.(19.35±0.34)μg/L,t=1.586,1.007,0.938,all P were>0.05)except for serum hyaluronidase[(1368.48±1093.82)U/L vs.(108.785±17.69)U/L,t=3.277,P<0.05).However,the serological indexes of rats in BDL 6W group were significantly higher than that in SO 6W group[AST:(599.00±98.34)U/L vs.(209.77±61.39)U/L;ALT:(165.03±110.58)U/L vs.(28.6±7.12)U/L;Serum hyaluronidase:(1690.43±619.15)μg/L vs.(135.99±16.87)μg/L;Serum typeⅣcollagen:(20.51±1.18)μg/L vs.(19.06±0.38)μg/L,t=5.845,3.442,4.928,3.754,all P<0.05].Liver Masson staining showed that both BDL4W and BDL 6W rats had obvious liver fibrosis changes.Compared with the rats in SO groups,the BDL6W
