摘　　要：目的探讨髓系来源抑制细胞(myeloid-derived suppressor cells,MDSC)在舌鳞状细胞癌(tongue squamous cell carcinoma,TSCC)患者外周血及肿瘤组织中的富集情况及对T细胞的免疫抑制功能研究。方法采集40例TSCC患者以及51例健康对照的外周血标本,利用流式细胞术检测外周血中MDSC的比例,并检测其对CD4+T与CD8+T细胞增殖功能的影响,进一步检测舌癌新鲜标本中MDSC的比例。结果与健康对照相比,TSCC患者外周血中的MDSC的比例显著升高,主要为多形核MDSC(polymorpho-nuclear-MDSC,PMN-MDS);该群细胞具有明显抑制T细胞增殖功能,此功能依赖于MDSC中活性氧(reac-tive oxygen species,ROS)的增加;肿瘤组织中MDSC比例高于癌旁组织相比(P<0.05),同样表现为粒细胞型。结论MDSC可能依赖于ROS的增加在舌癌相关肿瘤微环境中发挥重要的免疫抑制功能。Objective To explore the expression and functions of MDSC(myeloid-derived suppressor cell)and its immunosuppression for T cells in patients with TSCC(tongue squamous cell carcinoma).Methods Peripheral blood samples were collected from 40 patients with TSCC and 51 healthy controls.Flow cytometry was used to detect the proportion of MDSC in peripheral blood and its influence on the proliferation function of CD4+T and CD8+T cells,and the proportion of MDSC in fresh tongue carcinoma specimens was further detected.Results Compared with healthy controls,the proportion of MDSC in TSCC patients significantly increased,and majority of these cells were Polymorphonuclear-MDSC(PMN-MDS).These MDSCs were able to suppress the proliferation of T cell,which was dependent on the elevated reactive oxygen species(ROS)level.The proportion of MDSC in the tumor tissues also was higher than that in the adjacent tissues(P<0.05),which mainly were also PMN-MDSCs.Conclusion MDSC may play an important immunosuppressive role in tumor microenvironment of TSCC depending on ROS increasing.
