摘  要:目的:探讨TIAM1下调表达对食管癌(ESCA)细胞恶性增殖和迁移的影响。方法:采用免疫组织化学方法和RT-qPCR检测TIAM1在食管鳞癌临床组织中的表达情况;使用慢病毒介导的小干扰RNA构建TIAM1低表达的Eca109细胞株;CCK-8试验和集落形成测定评估TIAM1的表达对细胞增殖的影响。细胞划痕实验检测TIAM1对细胞迁移活性的影响;结果:相比于癌旁正常组织,ESCA组织中检测到的TIAM1表达水平更高(P<0.05)。下调TIAM1表达抑制了细胞增殖和集落形成(P<0.05),降低了细胞的迁移能力。结论:TIAM1在ESCA细胞的增殖和迁移中起着至关重要的作用,提示TIAM1可作为ESCA诊断及治疗中的潜在生物标志物。[Objective]To explore the effect of TIAM1 down-regulationon the malignant proliferation and migration of esophageal cancer cells.[Method]The expression patterns of TIAM1 in ESCA clinical tissues were evaluated using immunohistochemistry and RT-qPCR methods.Lentiviral-mediated small interfering RNAs were used to knockdown TIAM1 in Eca109 cell.CCK-8 and colony formation assays were used to assess the effect of TIAM1 on cell proliferation.The effect of TIAM1 on cell migratory activity was detected by cell scratch experiment.[Result]Higher level of TIAM1 expression was detected ESCA tissue compared to adjacent non-cancerous epithelium.Knockdown of TIAM1 expression was not only suppressed cell proliferation in CCK8 assay,colony formation assay(P<0.05),but also inhibited the cell migratory capacity in vitro(P<0.05).[Conclusion]TIAM1 played a critical role in ESCA cells proliferation and migration,suggesting TIAM1 to be a potential biomarker for the ESCA therapy.
