摘  要:目的了解开封市汉族育龄女性谷胱甘肽硫转移酶M1(GSTM1)、T1(GSTT1)和P1(GSTT1)基因多态性的分布特征。方法选取2018年3月至2019年12月在开封市妇幼保健院进行围孕期保健的506名汉族健康女性为研究对象。采集口腔黏膜上皮脱落细胞,提取基因组DNA。采用荧光定量PCR方法检测GSTM1、GSTT1和GSTP1基因多态性,进行统计分析。结果GSTM1和GSTT1缺失基因型频率分别为57.5%和47.0%。GSTP1 A313G基因位点突变纯合子GG基因型频率为3.2%,G等位基因频率为17.3%。GSTM1和GSTT1同时缺失基因型频率为28.5%,两个基因型的分布相互独立,无明显关联。结论获取开封市汉族育龄女性谷胱甘肽硫转移酶系相关基因多态性的群体遗传学特征,其中GSTM1和GSTT1相关基因位点缺失人群占比较高,这类人群对环境毒物的解毒能力较低,更应主动规避不良环境暴露,通过个体化营养干预来抵抗环境毒物带来的不良后果。Objective To understand the distribution characteristics of glutathione sulfur transferase M1(GSTM1),T1(GSTT1)and P1(GSTT1)gene polymorphisms in women of Han nationality in Kaifeng.Methods From March 2018 to December 2019,506 healthy Han females who underwent perinatal care in Kaifeng Maternal and Child Health Hospital were selected as the research subjects.Collected oral mucosal epithelial exfoliated cells and extracted genomic DNA.Fluorescence quantitative PCR method was used to detect GSTM1,GSTT1 and GSTP1 gene polymorphisms for statistical analysis.Results The frequencies of GSTM1 and GSTT1 deletion genotypes were 57.5% and 47.0% respectively.GSTP1 A313G locus mutation homozygous GG genotype frequency was 3.2%,G allele frequency was 17.3%.The frequency of simultaneous deletion of GSTM1 and GSTT1 was 28.5%,and the distribution of the two genotypes was independent of each other,with no obvious correlation.Conclusion The population genetic characteristics of glutathione thiotransferase-related gene polymorphism in Han-born women of childbearing age in Kaifeng are obtained.Among them,GSTM1 and GSTT1-related gene loci are missing,and the population has a low ability to detoxify environmental poison.In addition,it should take the initiative to avoid adverse environmental exposure,as well as individual nutrition intervention to resist the adverse consequences of environmental poisons.
