摘 要:目的探讨黑蒜多糖对X射线辐射所致小鼠免疫细胞和免疫因子损伤的保护作用。方法将100只雄性昆明小鼠按体重随机分为空白对照组、X线模型组、黑蒜多糖低、中、高剂量组(给药剂量分别为150、300、600 mg/kg·d)。黑蒜多糖各剂量组每日分别灌胃2 ml不同浓度的黑蒜多糖提取物,空白对照和X线模型组每日灌胃2 ml生理盐水,连续给药14 d后,除空白对照组外,使用直线加速器对各组动物进行全身X射线照射,6MV SSD 100 cm,处方剂量2Gy/d,剂量率400 cGy/min,吸收剂量百分比98.58%,照射时间28.5 s,连续照射2 d。辐射后24 h,分别检测小鼠外周血白细胞计数和分类、外周血T细胞亚群、血清IL-2和IFN-γ含量、骨髓DNA含量和细胞周期。结果与X线模型组比较,黑蒜多糖600 mg/kg剂量组白细胞计数增高,骨髓G1期细胞数量降低;300和600 mg/kg剂量组外周血淋巴细胞百分率增高,中性粒细胞百分率下降,血清IFN-γ含量升高,骨髓S期细胞数量升高;150、300、600 mg/kg剂量组血清IL-2含量增高,外周血CD3^+、CD4^、CD8^+T淋巴细胞比率升高。结论黑蒜多糖对X射线辐射引起的小鼠免疫功能损伤有一定的防护作用。Objective To study the protective effect of black garlic polysaccharides on X-ray induced immune injury in mice.Methods One hundred male Kunming mice were divided into control group,X-ray model group and black garlic polysaccharides low,medium and high dose group.Control group and X-ray model group were given 2m L of normal saline daily,while low,medium and high dose groups were given 150 mg/kg·d,300 mg/kg·d and 600 mg/kg·d of black garlic polysaccharides daily.After 14 days of administration,except for the control group,mice in each group were subjected to whole-body X-ray irradiation with a radiation dose rate of 400cGy/min,2Gy/d and 2d irradiation.White blood cell count and classification,serum IL-2 and IFN-γlevels,bone marrow DNA content,bone marrow cell cycle and peripheral blood T cell subsets were measured.Results Compared with the X-ray model group,the white blood cell count of the 600 mg/kg black garlic polysaccharides group increased and the G1 phase cells of the bone marrow decreased.In the 300 and 600 mg/kg groups,the ratio of peripheral blood lymphocytes increased,the ratio of neutrophils decreased,serum IFN-γlevels increased,and bone marrow S phase cells increased.Serum IL-2 was increased and the proportion of CD3+,CD4+and CD8+T lymphocytes in peripheral blood was increased in the 150,300 and 600 mg/kg groups.Conclusion Black garlic polysaccharides had a protective effect on mice immune function damage caused by X-ray radiation.
