摘  要:骨髓间充质干细胞(bone marrow mesenchymal stem cells,BM-MSCs)是一类重要的成体干细胞,具有形成成纤维细胞集落形成单位(CFU-F)的能力,并且能够分化为成骨细胞、软骨细胞和脂肪细胞。Foxp2属于Foxp蛋白家族,是一类保守的转录因子,在生物器官发育的多个过程中发挥重要作用。Foxp2基因最初被认为是一个与人类语言和认知能力调控相关的基因,有研究发现它的突变也会影响颅面骨的塑形和发育。因此本课题致力于研究Foxp2在MSC的增殖和成骨分化过程中的作用。结果表明,在骨髓间充质干细胞当中敲除Foxp2基因,导致干细胞增殖能力上升,而成骨分化能力下降;相反在间质细胞系C3H10T1/2和MC3T3当中过表达Foxp2,能抑制细胞的增殖,但可以促进其成骨分化。最后观察得出结论:Foxp2对MSC的成骨分化有促进作用,但会抑制MSC的增殖能力。这些研究结果将为理解颅骨与大脑组织的协同发育过程提供理论基础,也为骨质疏松的治疗提供线索。Bone marrow mesenchymal stem cells(BM-MSCs)is a critical adult stem cell population that have the capacity to form fibroblastic colony-forming units(CFU-f),and are able to differentiate into osteoblasts,chondrocytes and adipocytes.Foxp2,which belongs to the Fox protein family of transcription factors,plays important roles in multiple processes of biological development.Foxp2 was originally identified as a gene involved in language and cognition disorders in humans,and some studies have found that its mutation also affects the shaping and development of craniofacial bone.This project aims to study the role of Foxp2 in the replication and osteogenic differentiation of MSC.The knockout of Foxp2 gene in bone marrow mesenchymal stem cells resulted in increased proliferation of stem cells and decreased osteogenic differentiation.In contrast,Foxp2 overexpression in mesenchymal cell lines C3 H10 T1/2 and MC3 T3 inhibited cell proliferation but promoted osteogenic differentiation.In sum,it was concluded that Foxp2 promotes osteogenic differentiation but restrains the replicative capability of MSC.These results provide theoretical basis for understanding the co-variation process of skull and brain development,and also provide clues for the treatment of osteoporosis.
