摘  要:目的探讨电针刺激对单眼形觉剥夺弱视大鼠初级视皮层中多巴胺(dopamine,DA)、γ-氨基丁酸(gamma-aminobutyric acid,GABA)及其受体(D1受体和GABAA受体)mRNA表达的影响。方法选取36只鼠龄13 d的SD大鼠,随机分为3组正常对照组、弱视模型组、电针治疗组,每组12只。正常对照组大鼠不做任何处理,弱视模型组、电针治疗组大鼠右眼进行单眼形觉剥夺弱视造模。电针治疗组大鼠在生后30~60 d选取左侧太阳、合谷,右侧太阳、攒竹进行电针刺激干预,百会和右侧合谷仅给予针刺,不通电。每天同一时间段电针刺激干预30 min,频率2 Hz,脉冲长度0.1 s,每周至少干预6 d。采用高效液相色谱-紫外检测器和实时荧光定量PCR检测各组大鼠初级视皮层中DA、GABA含量以及D1受体、GABAA受体mRNA的表达情况。结果弱视模型组大鼠初级视皮层中DA、GABA的含量均低于正常对照组,差异均有统计学意义(均为P<0.05);电针治疗组DA的含量低于正常对照组,差异有统计学意义(P<0.05);而GABA的含量与正常对照组差异无统计学意义(P>0.05)。电针治疗组大鼠初级视皮层中DA、GABA的含量均高于弱视模型组,差异均有统计学意义(均为P<0.05)。弱视模型组和电针治疗组大鼠初级视皮层中D1受体、GABAA受体mRNA的表达均低于正常对照组,差异均有统计学意义(均为P<0.05);电针治疗组GABAA mRNA的表达高于弱视模型组,差异有统计学意义(P<0.05),而D1受体mRNA的表达与弱视模型组差异无统计学意义(P>0.05)。结论电针刺激会促进弱视大鼠初级视皮层中DA、GABA及其GABAA受体mRNA的表达。Objective To explore the effect of electroacupuncture(EA)on the mRNA expression of dopamine(DA),gamma-aminobutyric acid(GABA)and their receptor(D1,GABAA)mRNA in the primary visual cortex of rats with monocular deprived(MD)amblyopia.Methods Thirty-six SD rats aged 13 days were randomly divided into 3 groups normal control group,amblyopia model group and amblyopia+EA group,with 12 rats in each group.The rats in the normal control group did not do any treatment.The right eyes of the rats in the amblyopia model group and the amblyopia+EA group were modeled with monocular form deprivation amblyopia.Rats in the amblyopia+EA group were given electroacupuncture stimulation from 30 to 60 days after birth.The stimulating acupoints were left Taiyang,Hegu,right Taiyang,Zuanzhu,Baihui and right Hegu only given acupuncture without electricity.At the same time every day,electroacupuncture was intervened for 30 min with a frequency of 2 Hz and a pulse length of 0.1 s for at least 6 days per week.The expression of DA,GABA and their receptors’mRNA in the primary visual cortex were detected by high-performance liquid chromatography(HPLC)and quantitative polymerase chain reaction(Q-PCR)techniques.Results The contents of DA and GABA in the amblyopia model group were significantly lower than those in the normal control group,and the differences were both statistically significant(both P<0.05);the content of DA in the amblyopia+EA group was significantly lower than that in the normal control group(P<0.05),whereas there was no significant difference in content of GABA when compared with the normal control group(P>0.05).The contents of DA and GABA in the amblyopia+EA group were significantly higher than those in the amblyopia model group,and the differences were both statistically significant(all P<0.05).The expression of D1 and GABAA mRNA in the amblyopia model group and the amblyopia+EA group were significantly lower than those in the normal control group,and the differences were both statistically significant(all P<0.05).The express
