摘 要:目的测定不同壳聚糖的黏均分子量。方法采用乌氏毛细管黏度计法测定不同来源、不同脱乙酰度、不同灭菌方式及不同辐照剂量60Co灭菌壳聚糖的特性黏度,并利用经验公式推算黏均分子量。结果不同来源壳聚糖的相对分子质量相差较大,且与脱乙酰度成正相关;环氧乙烷及辐照灭菌均会使壳聚糖的相对分子质量下降,60Co辐照剂量为4~8 kGy时,壳聚糖相对分子质量降低幅度依次增大。结论乌氏毛细管黏度计法测定壳聚糖的黏均分子量简便、准确,不同处理方法对壳聚糖黏均分子量的影响较大。Objective To determine the viscosity average molecular weight of different chitosan.Methods The intrinsic viscosity of chitosan with different sources,different degrees of deacetylation,different ways of sterilization and different doses of 60 Co irradiation were determined by ubbelohde viscometer method,and the average molecular weight of viscosity was calculated by the empirical formula.Results The molecular weight of chitosan from different sources was quite different,and it was positively correlated with the degree of deacetylation.Ethylene oxide and irradiation sterilization would cause the molecular weight of chitosan to decrease.When the60 Co radiation dose increased from 4 kGy to 8 kGy,the relative molecular weight of chitosan decreased gradually.Conclusion The ubbelohde viscometer method is simple and accurate for determining the viscosity average molecular weight of chitosan.Different treatment methods have a great influence on the viscosity average molecular weight of chitosan.
