摘  要:目的探讨BimS过表达联合顺铂对涎腺腺样囊性癌(SACC)细胞株ACC-2增殖和凋亡的影响及分子机制。方法(1)体外培养ACC-2细胞,分别用浓度为0,2.5,5,10,20μg/ml的顺铂对呈现对数生长期的ACC-2细胞进行处理,处理后12,24,48 h采用MTT测定细胞增殖能力。(2)体外培养ACC-2细胞和BimS过表达的ACC-2细胞,分为对照组(control组)、顺铂(DDP)组、BimS过表达组、顺铂+BimS过表达组(BimS+DDP),对照组为未做任何处理的ACC-2细胞。四组细胞处理后均继续培养48 h,采用Western blot检测各组细胞Bax、caspase-3和Bcl-2蛋白相对表达水平;四组细胞接种至96孔板后,分别于12,24,48 h采用MTT检测各组细胞的增殖能力,流式细胞术检测各组细胞的凋亡情况。结果与0μg/ml相比,2.5,5,10,20μg/ml顺铂处理12,24,48 h细胞增殖抑制率均显著升高,呈剂量依赖性(P<0.01)。与DDP组和BimS组相比,DDP+BimS组12,24,48 h细胞增殖抑制率显著升高(P<0.01),细胞中Bcl-2蛋白表达显著减低,Bax、caspase-3蛋白表达显著升高(P<0.05),12,24,48 h细胞凋亡率显著升高(P<0.01)。结论BimS过表达联合顺铂可明显抑制ACC-2增殖,促进其凋亡,可能与二者联合作用能够下调Bcl-2蛋白、上调Bax蛋白,从而增强Bcl-2家族蛋白的促凋亡作用,活化下游caspase-3蛋白、激活线粒体凋亡途径有关。Objective To investigate the effects of BimS overexpression combined with cisplatin on proliferation and apoptosis of salivary adenoid cystic carcinoma cell line ACC-2.Methods①ACC-2 cells were cultured in vitro and treated with 0,2.5,5,10,20μg/ml cisplatin in the stage of logarithmic phase,respectively.After treatment for 12,24,48 h,the cell proliferation was measured by MTT assay.②ACC-2 cells and ACC-2 cells with overexpression of BimS were cultured in vitro and divided into control group,cisplatin group(DDP group),BimS overexpression group(BimS group)and BimS overexpression combined with cisplatin group(BimS+DDP group).ACC-2 cells were given no treatment in control group.After treatment,all the cells were continueously cultured for 48 h,and then Western blot analysis was used to evaluate the relative expression of Bax,caspase-3 and Bcl-2 proteins in each group.Cells in the four groups were inoculated to 96-well plates,and then the proliferation ability of cells in each group was measured by MTT,and the apoptosis of cells in each group was measured by flow cytometry at 12,24,and 48 h,respectively.Results Compared with 0μg/ml,the inhibition rates of cell proliferation were significantly increased after treated with 2.5,5,10 and 20μg/ml cisplatin at 12,24 and 48 h in a dose-dependent manner(P<0.01).Compared with BimS group and DDP group,the inhibition rate of cell proliferation was increased significantly at 12,24,48 h in BimS+DDP group(P<0.01).Compared with BimS group and DDP group,the expression of Bcl-2 proteins was significantly decreased in BimS+DDP group,while the expression of Bax and caspase-3 proteins were increased significantly(P<0.05).In addition,the apoptotic rate in BimS+DDP group was significantly higher than that in BimS group and DDPgroup at 12,24,48 h(P<0.01).Conclusion BimS overexpression combined with cisplatin can inhibit the proliferation of salivary adenoid cystic carcinoma cell line ACC-2 and promote its apoptosis.This effect may be mediated by the significant down-regulation of Bcl-
