摘 要:目的比较灵芝、枸杞、云芝、黄芪、香菇五种植物粗多糖对C57BL/6小鼠骨髓来源树突状细胞(bone marrow-derived dendritic cells,BMDC)的体外诱导功能。方法采用GM-CSF体外诱导和培养C57BL/6小鼠BMDC细胞,体外培养6 d后,分别加入灵芝、枸杞、云芝、黄芪和香菇五种植物粗多糖进行培养,24 h后,采用流式细胞术检测细胞表面CD80、CD86及MHCII分子的表达,采用ELISA检测BMDC细胞培养上清中细胞因子IL-6、IL-12和TFN-α的含量。结果云芝多糖可显著诱导BMDC细胞表明的CD80、CD86和MHCII分子表达上调,并刺激BMDC细胞释放IL-6,IL-12p40和TFN-α。枸杞多糖、灵芝多糖和香菇多糖仅显著性的诱导IL-6释放增加。结论云芝多糖对BMDC具有较强的诱导作用,具有潜在的疫苗佐剂活性。Objective To investigate the adjuvant activity of polysaccharides from Ganoderma lucidum,Lycium barbaru,Versicolor,Astragalus and Lentinan on bone marrow-derived dendritic cells(BMDCs)from C57 BL/6 mice.Methods Bone marrow cells from C57 BL/6 mice were cultured in the presence of granulocyte-macrophage colonystimulating factor GM-CSF.After 6 days,immature BMDCs were collected and co-cultured with the polysaccharides of G.lucidum,L.barbaru,Versicolor,Astragalus or Lentinula.Cp G oligodeoxynucleotides were used as a positive control.The cell surface expression of CD80,CD86 and MHCII were detected by fluorescence-activated cell sorting analysis.The release of cytokines interleukin(IL)-6,IL-12 and tumor necrosis factor TFN-αwere measured by enzyme-linked immunosorbent assay.Results Versicolor polysaccharides significantly upregulated the surface expression of CD80,CD86 and MHCII on BMDCs,as well as the release of the cytokines IL-6,IL-12 and TFN-αcompared with the control group.Conclusions Versicolor polysaccharides promoted the maturation of BMDCs in vitro and may have potential as an adjuvant for vaccine preparations.
