摘　　要：目的探讨长链非编码RNA(lncRNA)HOX转录反义RNA(HOTAIR)和lncRNA尿路上皮癌相关基因1(UCA1)在肾上腺恶性肿瘤中的表达及临床意义。方法收集2016年6月至2019年6月沧州市人民医院及沧州市中心医院收治的52例肾上腺癌患者手术切除的肾上腺癌组织及配对癌旁组织标本,提取RNA,反转录获得cDNA,采用实时荧光定量PCR检测癌组织和癌旁组织中lncRNA HOTAIR、lncRNA UCA1相对表达水平,分析其与临床病理特征的相关性。结果肾上腺癌组织中lncRNA HOTAIR、lncRNA UCA1相对表达水平显著高于癌旁正常组织(P<0.05);lncRNA HOTAIR相对表达水平与肿瘤最大径、远处转移、TNM分期有关(P<0.05),肿瘤最大径大、发生远处转移、TNM分期晚的患者lncRNA HOTAIR相对表达水平显著增高;lncRNA UCA1相对表达水平与肿瘤最大径、远处转移、包膜浸润、TNM分期有关(P<0.05),肿瘤最大径大、发生远处转移和包膜浸润、TNM分期晚的患者lncRNA UCA1相对表达水平显著增高。结论lncRNA HOTAIR、lncRNA UCA1在肾上腺癌组织中显著上调,其相对表达水平与肿瘤大小、转移和分期有关,可能成为肾上腺癌诊断和治疗新的分子标记物。Objective To investigate the expression and clinical significance of long-chain noncoding RNA(lncRNA)HOX transcript antisense RNA(HOTAIR)and lncRNA urothelial carcinoma associated 1(UCA1)in adrenal maligant tumor.Methods From June 2016 to June 2019,52 cases of adrenal carcinoma tissues and paired paracancerous tissues from Cangzhou People′s Hospital and Cangzhou Central Hospital were collected.RNA was extracted and retranscribed to obtain the cDNA.The relative expression levels of lncRNA HOTAIR and lncRNA UCA1 in cancer tissues and paracan cerous tissues was detected by real-time fluorescence quantitative PCR.The correlation between them and clinicopathological features was analyzed.Results The relative expressions levels of lncRNA HOTAIR and lncRNA UCA1 in adrenal carcinoma tissues were significantly higher than those in paracancerous normal tissues(P<0.05).The relative expression level of lncRNA HOTAIR was correlated with maximum diameter of tumor,distant metastasis and TNM stage(P<0.05).The relative expression level of lncRNA HOTAIR was significantly increased in patients with large maximum diameter of tumor,distant metastasis and late TNM stage.The relative expression level of lncRNA UCA1 was correlated with maximum diameter of tumor,distant metastasis,capsular infiltration and TNM stage(P<0.05).The relative expression level of lncRNA UCA1 was significantly increased in patients with large maximum diameter of tumor,distant metastasis and capsular infiltration,and late TNM stage.Conclusion LncRNA HOTAIR and lncRNA UCA1 are significantly up-regulated in adrenal carcinoma tissues,and their relative expression levels are related to the size,metastasis and stage of the tumors,which may provide new molecular markers for the diagnosis and treatment of adrenal carcinoma.
