摘  要:为了研究AA肉仔鸡肌肉组织中miR-133在不同饲养方式下的表达情况,试验采用生物信息学的方法分析了鸡肌肉组织中miR-133基因信息,比对了miR-133前体序列的同源性,通过stem-loop实时定量RT-PCR法分析gga-miR-133c-3p在不同饲养方式的AA肉仔鸡肌肉组织中的表达水平。试验结果表明,共有4个miR-133前体序列,分别位于鸡基因组23号、2号.3号和20号染色体上,4个miR-133的同源性可达96.59%;散养方式的AA肉仔鸡的翅肌、腿肌中gga-miR-133c-3p表达显著低于笼养方式,胸肌、心肌中gga-miR-133c-3p的表达显著高于笼养方式。In order to study the expression of miR-133 in muscle tissue of AA broilers under different feeding methods, the bioinformatics method was used to analyze the miR-133 gene information in chicken muscle tissues, and the homology of miR-133 precursor sequences was compared. The expression level of gga-miR-133c-3p in muscle tissues of AA broilers with different feeding methods was analyzed by real-time quantitative RT-PCR with stem-loop.The study results showed that there were four miR-133 precursor sequences, which were located on chromosomes 23, 2, 3, and 20 of the chicken genome. The homology of the four miR-133 could reach 96.59%. The expression of gga-miR-133c-3p in the wing muscles and leg muscles of AA broilers was significantly higher than that in cage culture. The expression of gga-miR-133c-3p in breast muscle and myocardium was significantly higher than that in cage culture.
