摘  要:目的探讨microRNA455(miR-455)在黑色素瘤中的表达及临床意义。方法通过实时荧光定量聚合酶链反应(qRT-PCR)法测定黑色素瘤组织和细胞系中miR-455的表达水平。结果与匹配的正常组织或正常表皮黑素细胞相比,黑色素瘤中miR-455显著下调,差异有显著性(P<0.05);另外,黑色素瘤细胞中miR-455表达增加,减少了细胞的增殖和侵袭(P<0.05)。结论在黑色素瘤中miR-455表达下调,并抑制黑色素瘤细胞的增殖和侵袭,miR-455的恢复可作为黑色素瘤患者的治疗方法。Objective To investigate the expression and significance of microRNA455(miR-455) in melanoma tissue and cell line.Methods In this study,the expression levels of miR-455 in melanoma tissues and cell lines were determined by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).Results Compared with matched normal tissues or normal epidermal melanocytes,miR-455 was significantly down-regulated in melanoma with a significant difference(P<0.05);in addition,increased expression of miR-455 in melanoma cells reduced cell proliferation and invasion(P<0.05).Conclusion These findings suggest that miR-455 is down-regulated in melanoma and inhibits the proliferation and invasion of melanoma cells.miR-455 recovery as a treatment for melanoma patients may be worth studying.
