血必净抑制大鼠角膜百草枯烧伤后新生血管形成的研究

摘  要:目的制作大鼠角膜百草枯烧伤导致角膜新生血管模型,观察在角膜新生血管(CNV)形成过程中核转录因子-κβ(NF-κβ)和血管内皮生长因子(VEGF)的动态表达,研究血必净注射液对NF-κβ和VEGF表达的影响以及抑制角膜新生血管形成的机制,为角膜百草枯烧伤的临床治疗奠定实验基础。方法将55只健康Wistar大鼠随机分成3组。A组:空白对照组5只;B组:实验对照组25只;C组:实验治疗组25只。制作大鼠角膜百草枯烧伤模型,B组每天腹腔注射0.9%生理盐水2ml,C组每天腹腔注射血必净注射液2ml,两组连续注射3周,并且每天氯霉素滴眼液点眼预防感染。第1,4,7,14及21天,观察角膜新生血管的生长情况,角膜做HE染色和免疫组织化学染色,检测NF-κβ和VEGF的动态表达,进行光密度值测定和分析。结果 HE染色:B组和C组第4天角膜基质出现新生血管,并于第14天数量达到顶峰,C组角膜新生血管数量始终低于B组,与B组比较,差异有显著性(P<0.01)。免疫组织化学染色:B组和C组角膜组织中NF-κβ和VEGF表达均上调,C组的NF-κβ和VEGF表达被抑制,与B组比较,差异有显著性(P<0.01)。结论 (1)大鼠角膜百草枯烧伤后,NF-κβ和VEGF的表达明显增加,NF-κβ和VEGF的高表达可能是角膜烧伤后形成新生血管的重要因素。(2)全身应用血必净注射液能抑制大鼠角膜NF-κβ和VEGF的表达。血必净注射液可能是由于抑制了某些炎性因子如NF-κβ和VEGF的表达,从而抑制角膜新生血管,达到治疗角膜百草枯烧伤的效果。Objective To observe the expression of VEGF and NF-κβ in cornea after paraquat burns,exploring the effect of Xuebijing injection on inhibition of CNV and the influence of the expression of VEGF and NF-κβ by establishing a model of corneal neovascularization induced by paraquat burn in rats.To explore the mechanism of Xuebijing injection on inhibition of CNV,and provide new direction of treating cornea burned with paraquat.Methods Fifty-five healthy Wistar rats were randomly divided into three groups:A blank control group(5 rats);B control group(25 rats);C experimental treatment group(25 rats).After paraquat-burning,intraperitoneal injection of 0.9% saline 2 ml and Xuebijing injection 2 ml were separatively injected into group B and C ereryday,stay three weeks,and dropped with Chloramphenicol Eye Drops every day to prevent infection.Observe corneal inflammation and growth of new blood vessels on the first,4 th,7 th,14 th,21 th days after operation,using HE staining and immunohistochemical staining to observe the corneal inflammatory reaction and the growth of new blood vessels and detect the expression of VEGF and NF-κβ,then measure and analysis optical density value with the computer image analysis system.Results HE staining:On day 4,neovascularization and inflammatory cells occurred in the corneal stroma in groups B and C,and peaked on the 14 th day.The number of corneal neovascularization in group C was always lower than that in group B.There was a significant difference between group B and group B(P<0.01).Immunohistochemical staining:the expression of VEGF and NF-κβ in groups B and C were raised.The expression of VEGF and NF-κβ were suppressed in group C,compared with group B there was significant difference(P<0.01).Conclusions(1)After corneal paraquat burns,the expression of VEGF and NF-κβ in cornea is obviously increase,high expression of VEGF and NF-κβ is likely to be an important factor in the formation of new blood vessels after corneal burns.(2)The general application of Xuebijing injection
