摘  要:目的研究单唾液酸四己糖神经节苷脂(GM1)对大鼠脑出血后神经功能障碍的作用及可能机制。方法通过自体血注入法建立大鼠尾状核脑出血模型,采用两种不同剂量(15 mg/kg、30 mg/kg)GMI进行干预,并将大鼠随机分为4组:假手术组,脑出血组,小剂量组,大剂量组。观察脑出血后大鼠神经功能评分,细胞凋亡情况,并采用Western印迹法检测各组脑组织多聚二磷酸腺苷核糖聚合酶(PARP)-1、凋亡诱导因子(AIF)、Bcl-2及Bax的表达量情况。结果脑出血后除假手术组外其余各组均出现不同程度的神经功能障碍,治疗组神经功能障碍轻于脑出血组,且大剂量组优于小剂量组(P<0.05);大鼠在脑出血后均出现细胞凋亡现象,治疗组细胞凋亡程度明显低于脑出血组,且存在量效关系(P<0.05);蛋白质印迹法检测结果显示:大鼠脑出血后均出现PARP-1、AIF表达上调,Bax/Bcl-2比值明显上升,经单唾液酸四己糖神经节苷脂治疗后PARP-1、AIF表达明显下调,而Bax/Bcl-2比值下降,且大剂量组变化程度大于小剂量组(P<0.05)。结论GMI在大鼠脑出血后的治疗中可能通过对PARP-1/AIF通路的调节,减轻脑出血后细胞凋亡程度,从而达到脑保护作用。Objective To study the effect of monosialotetrahexosyl ganglioside on cerebral hemorrhage in rats and its possible mechanism.Methods The rat model of caudate nucleus cerebral hemorrhage was established by autologous blood injection.Two different doses of monosialotetrahexosyl ganglioside(15 mg/kg,30 mg/kg)were used to intervene.The rats were randomly divided into four groups:sham group,ICH group,low dose group and high dose group.Neurological function score and apoptosis after intracerebral hemorrhage in rats were observed.Western blot was used to detect the expressions of PARP-1,AIF,Bax and Bcl-2.Results①All groups,except sham group,had different degrees of neurological dysfunction after intracerebral hemorrhage.The neurological dysfunction in monosialotetrahexosyl ganglioside group was milder than that in ICH group,and the high dose group was better than that of low dose group(P<0.05).②Apoptotic phenomena were observed in rats after intracerebral hemorrhage.The apoptotic degree of high dose group and low dose group were significantly lower than that of ICH group,and that of high dose group was lower than that of low dose group(P<0.05).③Western blot results showed that the expressions of PARP-1,AIF and Bax/Bcl-2 in ICH group were significantly higher than those in the two treatment groups(P<0.05),and the expressions of PARP-1,AIF and Bax/Bcl-2 were down-regulated after the intervention of monosialotetrahexosyl ganglioside(P<0.05),and the down-regulated degree in high dose group was greater than that in low dose group(P<0.05).Conclusions Monosialotetrahexosyl ganglioside plays a neuroprotective role in the treatment of cerebral hemorrhage in rats,and its mechanism is related to PARP-1/AIF pathway,which could reduce the apoptotic degree after cerebral hemorrhage and thus achieve cerebral protection.
