摘 要:牛病毒性腹泻病是危害养牛业的主要疾病之一。为了探讨牛病毒性腹泻病毒非编码区结构特征以及编码区突变规律。本文对病毒性腹泻JZ05-1毒株基因组序列进行5’、3’-UTR二级结构预测、毒力标记、结构蛋白编码区突变和RdRp同源建模等分析。研究发现JZ05-1毒株的5’-UTR在Ⅲa区与其他毒株结构具有明显差异,在150 nt左右区域形成1个独特的颈环结构,3’-UTR可分为3个独立的结构区域,其他参考毒株不具备此特征;结构蛋白编码区分析,JZ05-1毒株具有大量的独特碱基,特有碱基位点有201处;非结构蛋白区域分析,差异氨基酸主要存在于RdRp蛋白表面可及性好的区域,且多位于疏水区,蛋白内部活性中心区域的氨基酸位点极少突变;毒力标记分析,278位做为毒力标记的规律不适用于JZ05-1毒株。结果表明,JZ05-1毒株与其他同型毒株有明显差异。本研究可为疫苗的研制提供理论基础。Bovine viral diarrheal disease is one of the serious diseases caused by bovine viral diarrheal virus,which has a worldwide distribution.This study of viral diarrhea JZ05-1 strain genome sequence 5’,3’UTR secondary structure prediction,virulence markers,structural protein coding region mutations and RdRp homology modeling were analyzed.The results showed that the structure of 5’-UTR inⅢarea in JZ05-1 strain was significantly different with in other strains,and a unique neck ring structure was formed in about 150 nt region.3’-UTR can be divided into three independent structural regions,and other reference strains did not have this feature.The structural protein coding region analysis showed that JZ05-1 strain had a large number of unique bases,and the unique base site was 201.According to the analysis of non-structural protein region,the difference amino acids mainly existed in the regions with good surface accessibility on the surface of RdRp protein,and most of them were located in hydrophobic region,and the amino acid sites in the active center regions of the protein were rarely mutated.Virulence marker analysis showed that the rule of 278 as a virulence marker was not suitable for JZ05-1 strain.The above results showed that the JZ05-1 strain was significantly different from other strains,which provided a theoretical basis of the development of vaccine.
