摘　　要：目的研究组蛋白去乙酰化酶抑制剂Givinostat对三阴性乳腺癌细胞的作用及其机制。方法采用MTT法,细胞划痕实验,Transwell小室实验,Western blot实验,流式细胞分选实验检测Givinostat抑制剂对三阴性乳腺癌细胞的作用。结果 Givinostat抑制三阴性乳腺癌增殖并呈浓度和时间依赖性(P<0.05);抑制细胞迁徙和侵袭(P<0.05);通过下调Bcl-2、Oct4、C-myc蛋白表达,上调Bax、Caspase-3蛋白促进细胞凋亡(P<0.05);对表达CD44+CD24-的乳腺癌干细胞数量无影响(P>0.05)。结论 Givinostat可以抑制三阴性乳腺癌细胞增殖、迁徙和侵袭,并且促进凋亡及其相关蛋白的表达,对干细胞蛋白Oct4、C-myc表达有抑制作用。Objective To study the effect and mechanism of histone deacetylase inhibitor: Givinostat on triple negative breast cancer cells. Methods MTT method, cell scratch test, Transwell cell test, Western blot test and flow cytometry test were used to detect the effect of Givinostat inhibitor on triple negative breast cancer cells. Results Givinostat inhibited the proliferation of triple-negative breast cancer in a concentration-and time-dependent manner(P<0.05);inhibiting cell migration and invasion(P<0.05);down-regulating the expression of Bcl-2, Oct4, and C-myc proteins, and up-regulating Bax and Caspase-3 protein to promotes apoptosis(P<0.05);it had no effect on the number of breast cancer stem cells expressing CD44^+CD24^-(P>0.05). Conclusion Givinostat can inhibit the proliferation, migration and invasion of triple-negative breast cancer cells, promote the expression of apoptosis and relate proteins, and inhibit the expression of stem cell proteins Oct4 and C-myc.
