摘 要:目的探究兔VX2骨肿瘤模型中干扰素(interferon,IFN)-γ、白介素(interleukin,IL)-4和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的表达水平及其相关性。方法30只日本大耳白兔随机分为实验组和对照组,各15只,两组植瘤2周后抽取外周血并处死,采用酶联免疫吸附试验法检测外周血IFN-γ、IL-4及肿瘤组织IFN-γ、IL-4和PCNA的表达水平并留取病理组织检测。结果与对照组比较,实验组外周血及肿瘤组织IFN-γ的表达水平降低(P<0.05),IL-4的表达水平上升(P<0.05);肿瘤组织PCNA的表达水平上升(P<0.05)。实验组外周血与肿瘤组织的IFN-γ水平呈正相关(r=0.737,P=0.002),外周血与肿瘤组织的IL-4水平呈正相关(r=0.869,P<0.01);外周血及肿瘤组织IFN-γ与IL-4的水平呈负相关(r=-0.537,P=0.039;r=-0.812,P<0.01)。实验组肿瘤组织PCNA与外周血及肿瘤组织IFN-γ呈负相关(r=-0.604,P=0.017;r=-0.785,P=0.001),与IL-4呈正相关(r=0.884,P<0.01;r=0.846,P<0.01)。结论种植VX2肿瘤后肿瘤增殖迅速,免疫调节受到抑制,表现为IL-4和PCNA表达水平上升,IFN-γ表达水平下降,IFN-γ与IL-4、PCNA呈负相关,IL-4与PCNA呈正相关。Objective To investigate the expression levels of interferon(IFN)-γ,interleukin(IL)-4 and proliferating cell nuclear antigen(PCNA)and their correlation in a rabbit VX2 bone tumor model.MethodsThirty Japanese white rabbits were randomly divided into an experimental group and a control group(15 rabbits each).After 2 weeks of tumor implantation in both groups,peripheral blood was extracted and sacrificed.The enzyme-linked immunosorbent assay was used to detect the expression level of peripheral blood IFN-γ,IL-4 and tumor tissue IFN-γ,IL-4 and PCNA and examined by pathological tissues.ResultsCompared with the control group,the expression level of IFN-γin the peripheral blood and tumor tissue of the experimental group decreased(P<0.05),and the expression level of IL-4 increased(P<0.05),and the expression level of PCNA in tumor tissue increased(P<0.05).In the experimental group,the levels of IFN-γin peripheral blood and tumor tissues were positively correlated(r=0.737,P=0.002),and peripheral blood was positively correlated with IL-4 levels in tumor tissue(r=0.869,P<0.01),and peripheral blood and tumor tissue IFN-γwas negatively correlated with IL-4 levels(r=-0.537,P=0.039;r=-0.812,P<0.01).In the experimental group,tumor tissue PCNA was negatively correlated with peripheral blood and tumor tissue IFN-γ(r=-0.604,P=0.017;r=-0.785,P=0.001),which was positively correlated with IL-4(r=0.884,P<0.01;r=0.846,P<0.01).ConclusionAfter implantation of VX2 tumors,tumor proliferation was rapid and immune regulation was suppressed,which showed increased expression levels of IL-4 and PCNA and decreased expression levels of IFN-γ.IFN-γwas negatively correlated with IL-4 and PCNA,and IL-4 was positively correlated with PCNA.
