摘  要:目的探究黄芩苷(Baicalin)抑制NF-κB p65的活化对乙型病毒性肝炎大鼠模型肝功能损伤的调节作用。方法选取90只SD大鼠,并分为健康对照组,乙肝病毒模型组、阳性对照(拉米夫定)组、黄芩苷25umol/L组、黄芩苷50umol/L组、黄芩苷100umol/L组各15只;阳性对照使用拉米夫定10 mg/kg处理,加药组分别采用对应剂量的黄芩苷处理,乙肝病毒模型组采用等剂量的生理盐水处理。检测各组大鼠ALT、AST、TC、TG、HDL及LDL含量;HE染色观察各组大鼠肝组织结构排列及损失情况;检测各组大鼠Caspase-3、Survivin的表达情况;油红O染色检测脂肪堆积情况;检测各组大鼠外周血中iNOS、IL-6、IL-10、IL-4的水平;使用免疫组化和Western blot检测NF-κB p65的核定位和核表达情况;分别加入大黄素和细菌内毒素脂多糖(LPS)处理大鼠,分别检测上述指标。结果相比健康对照组,乙肝病毒模型组、黄芩苷25umol/L组、黄芩苷50umol/L组AST、ALT、Caspase-3基因及蛋白表达、TC、TG、HDL、iNOS、IL-6水平显著升高,Survivin基因及蛋白表达、HDL水平显著降低(P<0.05);相比健康对照组,乙肝病毒模型组和黄芩苷25umol/L组IL-10、IL-4的水平显著降低,黄芩苷100umol/L组IL-10、IL-4的水平显著升高(P<0.05);相比乙肝病毒模型组,拉米夫定组、黄芩苷25umol/L组和黄芩苷50umol/L组AST、ALT、Caspase-3基因及蛋白表达、TC、TG、HDL、iNOS、IL-6水平显著降低,Survivin基因及蛋白表达、HDL、IL-10、IL-4水平显著升高(P<0.05);相比LPS组,LPS+黄芩苷100um组NF-κB p65蛋白表达水平、AST、ALT、iNOS显著降低,IL-10显著升高(P<0.05)。结论黄芩苷可以通过抑制NF-κB p65的活化并降低炎症反应、抑制肝细胞的凋亡实现调节乙型病毒性肝炎大鼠肝脏功能。To explore effect of Baicalin on liver damage in rat models of hepatitis B virus by inhibiting NF-κB p65 activation, 90 SD rats were enrolled and divided into healthy control group, hepatitis B virus(HBV) model group, positive control group, Baicalin 25 umol/L, 50 umol/L and 100 umol/L group, with 15 cases in each group.The positive control group was treated with 10 mg/kg lamivudine;the medication groups were respectively treated with corresponding dose of Baicalin;while the HBV model group was treated with equal dose of normal saline. The contents of ALT, AST, TC, TG, HDL and LDL were detected in each group;the structure arrangement and damage of liver tissues were observed by HE staining in each group;the expression of Caspase-3 and survivin was detected in each group. Oil red O staining was applied to detect fat accumulation. The levels of iNOS, IL-6, IL-10 and IL-4 in peripheral blood were detected. The immunohistochemistry and Western blot were applied to detect nuclear localization and nuclear expression of NF-κB p65. The rats were treated with emodin and endotoxin lipopolysaccharide(LPS), all the above-mentioned indexes were detected. Data showed that as compared with healthy control group, the gene and protein expression of AST, ALT, Caspase-3, the levels of TC, TG, HDL, iNOS and IL-6 were significantly increased in HBV model group, Baicalin 25 umol/L group and Baicalin 50 umol/L group, while Survivin gene and protein expression, and HDL level were significantly decreased(P<0.05).Compared with healthy control group, the levels of IL-10 and IL-4 were significantly decreased in HBV model group and Baicalin 25 umol/L group, while the levels of IL-10 and IL-4 were significantly increased in Baicalin 100 umol/L group(P<0.05).
