摘  要:目的采用生物信息分析方法探讨RUNT相关转录因子1(RUNX1)在肠癌中的表达及其与患者预后的关系。方法采用生物信息分析方法分析肿瘤基因组图谱(TCGA)数据库中RUNX1基因的表达(mRNA)情况。应用蛋白-蛋白作用网络数据库STRING构建RUNX1基因编码蛋白相互作用网络,对蛋白网络中的基因进行基因本体论(GO)和京都基因及基因组百科全书(KEGG)分析,预测其可能功能及信号通路。根据RUNX1基因在肿瘤组织中表达的中位数分为高和低表达组,绘制生存曲线,生分析探讨RUNX1高低表达水平与肠癌的预后关系。结果RUNX1在结直肠癌组织中的表达水平明显高于正常肠黏膜组织(P<0.05)。RUNX1编码蛋白及相互作用蛋白主要定位于细胞核、细胞膜和染色质,分子功能主要富集于蛋白结合、细胞核蛋白结合及蛋白复合体。而生物学过程主要富集于细胞代谢,细胞增殖和细胞对外界刺激反应。KEEG信号通路分析显示RUNX1及相关基因主要通路富集于消化系统肿瘤、磷酸肌醇3激酶(PI3K)、细胞周期和Wnt等肿瘤相互信号通路。Log-rank检验显示RUNX1高、低表达组总生存[风险比(HR)=1.5,P>0.05]差异无统计学意义而高表达组无疾病进展生存(HR=1.9,P<0.01)显著低于低表达组。亚组分析显示,结肠癌RUNX1高表达者总生存(HR=1.7,P<0.05)和无疾病进展生存(HR=1.9,P<0.01)均低于低表达者。而直肠癌中RUNXI1高低表达对患者预后无明显影响(P>0.05)。结论在结肠癌和直肠癌中,RUNX1基因表达水平明显上调并与结肠癌患者的预后不良有关。Objective To investigate the expression of RUNT-like transcription factor 1(RUNX1)in colorectal cancer and its relationship with prognosis by bioinformatics analysis.Methods Bioinformatics analysis was used to analyze the expression of RUNT gene in the cancer genome atlas(TCGA)database.Using protein-protein interaction network data,STRING constructed RUNX1 protein-coding interaction network.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)were used to analyze the genes in protein network and predict their possible functions and signaling pathways.According to the median expression of RUNX1 gene in cancer tissues,the patients were divided into high and low expression groups.Survival curve was drawn to analyze the relationship between the expression of RUNX1 and the prognosis of colorectal cancer.Results The expression level of RUNX1 in colorectal cancer tissues was significantly higher than that in normal mucosa tissues(P<0.05).RUNX1 encoding proteins and interacting proteins are mainly located in nucleus,cell membrane and chromatin,and their molecular functions are mainly concentrated in protein binding,nuclear protein binding and protein complexes.Biological processes are mainly concentrated in cell metabolism,cell proliferation and cell response to external stimuli.KEGG signaling pathway analysis showed that RUNX1 and related genes were mainly involved in the interaction signaling pathways of digestive system tumors,phosphatidylinositol 3 kinase(PI3K),cell cycle and Wnt.Log-rank test showed that there was no significant difference in total survival[hazard ratio(HR)=1.5,P>0.05]between the high and low expression groups of RUNX1,while the disease-free progressive survival(HR=1.9,P<0.005)in the high expression group was significantly lower than that in the low expression group.Subgroup analysis showed that the total survival(HR=1.7,P<0.05)and disease-free progression survival(HR=1.9,P<0.01)of patients with high RUNX1 expression were lower than those with low RUNX1 expression.The expression of RUNX
