摘  要:为了研究抗生素抗性菌、抗性基因的扩散和污染对环境和人类健康可能产生的威胁,分8次采集城市污水处理厂水样,应用肉汤微量稀释法和聚合酶链式反应方法,分离、鉴定对四环素、磺胺甲恶唑敏感菌株,分析其抗生素抗性,检测抗性基因。从某污水处理厂出水中分离22株四环素类抗性菌、26株磺胺类抗性菌,检测其对常用抗生素耐受性及抗生素浓度、抗性基因含量。测得四环素类和磺胺类双重抗性菌数量为5.04×102~2.51×103 CFU/mL;抗性菌对9种抗生素具有抗性,最高耐受力为512 mg/mL,其中,对氯霉素耐受能力最强,对青霉素、氨苄青霉素、头孢氨苄、环丙沙星、庆大霉素、阿奇霉素耐受力次之,对利福平耐受力最弱;还检测到双重抗性基因,其中tetW和sul1的含量较高。结果表明:城市污水处理厂的四环素类和磺胺类抗生素含量较高,存在双重抗性菌、抗性基因。In order to study the antibiotic-resistant bacteria,its spread and pollution of resistance genes may pose threats to the environment and human health.The water samples were collect from municipal sewage treatment plants by 8 times.The broth microdilution method and polymerase chain reaction method was used to separate and identify of tetracycline,sulfamethamine oxazole-sensitive strains,and their antibiotic resistance,and detected resistance genes.The following results are got from the research.22 tetracycline-resistant bacteria and 26 sulfonamide resistant bacteria are isolated from the effluent of a sewage treatment plant.Their tolerance to common antibiotics,tetracycline antibiotics,and antibiotic resistance genes(ARGs)were tested.The tetracycline and sulfonamide double resistant bacteria are in 5.04×102~2.51×103 CFU/mL.The separated sulfonamides resistant bacteria also exhibits resistance to other 9 kinds of antibiotics.And so antibiotic tolerance is the highest of 512 mg/L,sulfonamides resistant bacteria has the highest resistance to chloramphenicol,middle resistance to tolerance ability of penicillin,ampicillin,cephalothin,and the lowest resistance to rifamicin.In the collected samples,the highest relative abundances are SUL1 and tetW.All these results show that the content of tetracyclines and sulfonamides in municipal wastewater treatment plants is high,and there are dual resistant bacteria and resistance genes.
