摘 要:目的:分析miR-372通过程序性细胞死亡4(PDCD4)调节白血病细胞U937增殖和凋亡的机制。方法:白血病细胞U937分别转染miR-372 inhibitor(Anti-miR-372组)、inhibitor control(Anti-NC组)、PDCD4 siRNA和miR-372 inhibitor(Anti-miR-372+si-PDCD4组)、siRNA control和miR-372 inhibitor(Anti-miR-372+si-NC组),未行转染的常规培养U937细胞为Control组,CCK-8法测定各组细胞增殖,流式细胞术测定各组细胞凋亡,Western blot测定各组细胞中Bax、Bcl-2蛋白表达。生物信息数据库预测miR-372和PDCD4靶向关系,荧光素酶报告系统鉴定二者靶向关系。结果:与Anti-NC组比较,Anti-miR-372组白血病细胞U937增殖能力降低,凋亡率升高,细胞中Bax蛋白表达水平升高,Bcl-2蛋白表达水平降低(P均<0.01)。miR-372和PDCD4互为靶向关系。PDCD4 siRNA转染可以提高下调miR-372的白血病细胞U937增殖能力,减少细胞凋亡,促进细胞中Bcl-2蛋白表达,减少细胞中Bax蛋白表达水平(P均<0.01)。结论:下调miR-372通过PDCD4抑制白血病细胞U937增殖并诱导细胞凋亡。Objective:To explore the mechanism of miR-372 through programmde cell death 4(PDCD4)to regulate the proliferation and apoptosis of leukemia cells U937.Method:The leukemia cells U937 were transfected with miR-372 inhibitor(Anti-miR-372 group),inhibitor control(Anti-NC group),PDCD4 siRNA and miR-372 inhibitor(Anti-miR-372+si-PDCD4 group),siRNA control and miR-372 inhibitor(Anti-miR-372+si-NC group).Cell proliferation was measured by CCK-8 method,and cell apoptosis was measured by flow cytometry,the expression of Bax and Bcl-2 protein in each cell was measured by Western blot.The biological information database predicts the targeting relationship between miR-372 and PDCD4,and the luciferase reporting system identifies the targeting relationship between the two.Results:Compared with the Anti-NC group,the Anti-miR-372 group's leukemia cells had reduced U937 proliferation ability,increased apoptosis rate,increased Bax protein expression level,and reduced Bcl-2 protein expression level(P all<0.01).miR-372 and PDCD4 were targeted to each other.PDCD4 siRNA transfection could increase the proliferation ability of U937 leukemia cells that down-regulate miR-372,reduce apoptosis,promote Bcl-2 protein expression in cells,and reduce Bax protein expression levels in cells(P all<0.01).Conclusion:Down-regulation of miR-372 can inhibit the proliferation of U937 leukemia cells and induce apoptosis through PDCD4.
