摘 要:目的研究不同月龄精神分裂症模型小鼠的前脉冲抑制(prepulse inhibition,PPI)改变。方法将小鼠按照月龄分为青年、中年和老年组,按小清蛋白(parvalbumin,PV)阳性的γ-氨基丁酸能神经元中精神分裂症风险基因Erbb4基因型分为精神分裂症模型(PV-Erbb4-/-)小鼠和正常对照(PV-Erbb4+/+)小鼠,用前脉冲抑制实验检测PPI效率(PPI%),前脉冲刺激为74 dB、78 dB和86 dB,用一般线性回归分析Erbb4基因型和年龄对PPI%的交互作用。结果前脉冲刺激为78 d B和86 dB时,Erbb4基因型和年龄对PPI强度的交互作用有统计学意义(P<0.05),正常对照(PV-Erbb4+/+)小鼠老年组PPI%比青年组更小(78 dB:β=-13.61,P=0.02;86 dB:β=-10.58,P=0.03)。结论年龄和Erbb4基因型对PPI的影响存在交互作用,正常对照小鼠PPI随年龄增加而减小,而精神分裂症模型小鼠变化方向相反。Objective To study the differences of prepulse inhibition(PPI)of schizophrenia model mice in three different age groups.Methods Mice with specific depletion of Erbb4 in parvalbumin(PV)-expressing GABAergic interneurons were used as schizophrenia model mice(PV-Erbb4-/-)and mice without depletion(PV-Erbb4+/+)as healthy control.All mice were divided into three age groups,adult group,middle-aged group and geriatric group.PPI test was conducted using three different prepulse stimulus 74 dB,78 dB,and 86 dB.The interaction of Erbb4 genotype and age on PPI%was then tested using linear regression model.Results There was significant interaction between Erbb4 genotype and age when the prepulse stimulus was 78 dB and 86 dB(P<0.05).In healthy control mice(PV-Erbb4+/+),PPI%was lower in geriatric group than in adult group(78 dB:β=-13.61,P=0.02;86 dB:β=-10.58,P=0.03).Conclusion There is an interaction between Erbb4 genotype and age on the effect on PPI.PPI decreases with age in healthy control mice(PV-Erbb4+/+).
