摘  要:目的探讨加味温胆汤对慢性不可预见性温和应激(chronic unpredictable mild stress,CUMS)抑郁模型大鼠的抗抑郁作用及色氨酸—犬脲氨酸(Tryptophan-kynurenine,TRP-KYN)代谢通路的影响。方法将48只SD大鼠按照随机数字表法随机分为四组,每组12只,分别为空白组、模型组、氟西汀对照组(简称西药组)、加味温胆汤组(简称中药组);除空白组外,其余各组进行21天孤养加CUMS方法复制大鼠抑郁症模型。模成后西药组给予盐酸氟西汀1.8 mg/(kg·d),中药组给予加味温胆汤12 g/(kg·d),每日1次,连续灌胃4周。运用旷场实验、糖水偏好实验评价各组大鼠的抑郁行为,采用ELISA法检测血清TRP-KYN代谢通路相关分子含量。结果加味温胆汤能增加CUMS抑郁大鼠的糖水偏爱率,提高旷场实验得分,具有显著的抗抑郁效应。和模型组比较,中药组和西药组能显著提高血清TRP、KYNA含量(P<0.05)、显著降低脲氨酸(3-Hydroxy kynurenine,3-HKYN)、3-羟邻氨苯甲酸(3-Hydroxy anthranilic acid,3-HAA)、喹啉酸(Quinolinic acid,QUIN)、KYN含量(P<0.05),中药组和西药组之间差异无明显差异。加味温胆汤能降低KYN/TRP、KYN/KYNA比值,升高KYN/QUIN比值,和模型组相比差异具有统计学意义(P<0.05)。结论加味温胆汤可以改善CUMS大鼠的抑郁行为具有抗抑郁效应,能调节TRP-KYN代谢通路,抑制神经毒性,促进神经保护代谢的作用。Objective To investigate the antidepressant effect of Jiawei Wendan decoction(JWD)and the effect of TRP-KYN metabolic pathway on depression model rats of CUMS.Methods Forty-eight SD rats were randomly divided into four groups with 12 rats in each group:blank group,model group,fluoxetine control group and modified Jiawei Wendan decoction group.Fluoxetine hydrochloride 1.8 mg/(kg·d).Depressive behavior was evaluated by open field test and sugar preference test,and serum TRP-KYN metabolic pathway related molecules were detected by ELISA.Results Jiawei Wendan decoction could increase the sugar preference rate and open field test score of CUMS depression rats,and had significant antidepressant effect.Compared with model group,TCM group and Western medicine group could significantly increase the content of serum TRP,KYNA(P<0.05),significantly reduce the content of KYN,3-HKYN,3-HAA,QUIN(P<0.05),and there was no significant difference between TCM group and Western medicine group.Modified Jiawei Wendan decoction could decrease the ratio of NKY/TRP,KYN/KYNA,and increase the ratio of KYN/QUIN,which was significantly different from the model group(P<0.05).Conclusion Jiawei Wendan decoction can improve the depressive behavior of CUMS rats with antidepressant effect,regulate TRP-KYN metabolic pathway,inhibit neurotoxicity and promote neuroprotective metabolism.
