摘  要:对植物乳杆菌、罗伊氏乳杆菌和鼠李糖乳杆菌防御鼠伤寒沙门菌作用进行了比较研究。通过将此3株乳酸菌菌株灌胃SPF级C57BL/6小鼠后,感染鼠伤寒沙门菌,通过监测小鼠体重及存活率、肠道病理组织学观察、沙门菌器官荷菌数及肠黏膜SIgA的分泌初步评价其抗沙门菌效应;利用Illumina MiSeq测序平台对小鼠盲肠菌群多样性进行高通量测序分析。结果显示,植物乳杆菌与鼠李糖乳杆菌干预后,均能抑制沙门菌引起的小鼠体重下降,提高小鼠存活率,缓解沙门菌引起的肠道损伤,抑制沙门菌向肝脏、脾脏及肠系膜淋巴结的侵袭;植物乳杆菌的作用效果更为显著;菌群多样性分析表明,植物乳杆菌与鼠李糖乳杆菌均能明显改善沙门菌感染后引起的菌群紊乱,恢复厚壁菌门、拟杆菌门等优势菌群与变形菌门之间的平衡,且能够显著提高厚壁菌门中乳酸杆菌属、毛螺旋菌属等有益菌数量,降低变形菌门中志贺氏菌、沙门菌等致病菌数量,而罗伊氏乳杆菌对沙门氏菌感染几乎无保护作用。表明植物乳杆菌对鼠伤寒沙门菌感染具有较好的防御作用。Salmonella is a kind of pathogenic bacterial species which caused bacterial foodborne zoonosis in humans.Probiotics has prominent advantage on prevention and treatment of intestinal pathogenic bacterial infections.In this study,several Lactobacillus strains were orally administrated to the mice respectively,following with Salmonella typhimurium infection.The defensive effect of Lactobacillus strains were evaluated and compared by detecting body weight,survival rate,histopathology analysis,Salmonella loads in tissues,and SIgA secretion after challenge with Salmonella.The diversity of intestinal flora was also sequenced and analyzed by Illumina MiSeq platform.The result showed that L.plantarum could better inhibit body weight loss caused by Salmonella infection,and increase the survival rate,which was similar to the mice treated with LGG.Intestinal inflammation induced by Salmonella was also relieved significantly in mice treated with L.plantarum or LGG.In addition,L.plantarum inhibited Salmonella transloctation into tissue and also promoted more SIgA secretion than that of LGG.Analysis of intestinal flora diversity displayed that L.plantarum and LGG could restore the balance among Firmicutes,Bacteroidetes and Proteobacteria,which could be broken in mice challenged with Salmonella,to display defensive effect against Salmonella.Interestingly,L.plantarum and LGG increased population of beneficial flora including Lachnospiraceae and Lactobacillus belonged to Firmicutes,decreased population of pathogenic flora including Escherichia,Shigella and Salmonella belonged to Proteobacteria.L.reuteri did not have effect of defense against Salmonella.To conclude,L.plantarum had better effect of defense against Salmonella,which cloud be used to further study on beneficial effect and mechanism.
