摘 要:目的观察绿茶在川芎茶调散治疗偏头痛的配伍使用中是否具有协同增效作用,及其潜在的作用机制。方法选用颈背部皮下注射硝酸甘油构建偏头痛大鼠模型,观察各组给药后挠头、爬笼及洗脸次数等行为学评价指标变化,并检测大鼠血清中5-羟色胺(5-HT)和β-内啡肽(β-EP)的含量变化。结果与正常组相比,模型组大鼠在各时间段行为学评分均显著增加(P<0.01);且血清中5-HT、β-EP均显著降低(P<0.01),证明模型构建成功。川芎茶调散组、绿茶组、川芎茶调散(去绿茶)组在给药后30~90min内行为学评分与模型组相比均显著下降(P<0.05),三者任2组间无显著性差异。川芎茶调散组、川芎茶调散(去绿茶)组和茶碱组血清中5-HT含量与模型组比均显著升高(P<0.05),三者任2组间无显著性差异,而绿茶组和茶多酚组血清中5-HT含量与模型组相比无显著性差异。川芎茶调散组、川芎茶调散(去绿茶)组血清中β-EP含量与模型组相比均显著升高(P<0.01),两者之间无显著性差异;茶碱组和茶多酚组血清中β-EP含量与模型组相比均显著降低(P<0.01);绿茶组、茶碱组和茶多酚组与川芎茶调散组比,血清中β-EP含量均显著降低(P<0.01)。结论绿茶对川芎茶调散治疗偏头痛没有明显的协同增效作用。其机理可能是绿茶和茶碱、茶多酚促进了血清中5-HT的释放,但是茶碱和茶多酚却抑制了β-EP的释放,起到了一定的消减作用,所以茶碱和茶多酚不是绿茶止痛的主要药效物质。本研究为川芎茶调散的临床合理应用及其现代化研究提供了实验依据。Objective: To observe the synergistic effect of green tea in the compatibility of Chuanxiong Chatiao San in the treatment of migraine patients and its potential action mechanism. Methods: The migraine rat model was made by subcutaneous injection of nitroglycerin on the back of the neck. The changes in behavioral evaluation indexes as head scratching, cage climbing and face washing in each group after administration were observed, and the serotonin(5-HT)and β-endorphin(β-EP)content changes in the rat serum was detected. Results: Compared with the normal group, the behavioral scores of the rats in the model group increased significantly at each time period( P <0.01)and 5-HT and β-EP in the serum significantly decreased( P <0.01), which proved the model building was successful. The behavioral scores of the Chuanxiong Chatiao San group, the green tea group, and the Chuanxiong Chatiao San(de-green tea)group were significantly lower than that of the model group within 30 to 90 minutes after administration( P <0.05). There was no significant difference between any two groups of the three. The serum 5-HT content in the Chuanxiong Chatiao San group, Chuanxiong Chatiao San(de-green tea)group and theophylline group were significantly higher than that in the model group( P <0.05), and there was no significant difference between any two groups of the three. There was no significant difference in serum 5-HT content between the green tea group and the tea polyphenol group when compared with the model group. Compared with the model group, the serum β-EP content in the Chuanxiong Chatiao San group and Chuanxiong Chatiao San(de-green tea)group were significantly increased( P <0.01), and there was no significant difference between the two. The serum β-EP content of the theophylline group and tea polyphenol group was significantly lower than that of the model group( P <0.01).
