摘 要:目的:研究过表达脆性组氨酸三联基因(FHIT)对食管鳞癌KYSE150细胞增殖和凋亡的影响及可能的作用机制。方法:实时荧光定量PCR(qRT-PCR)法检测59例食管鳞癌和对应的癌旁正常组织中FHIT mRNA表达水平。将KYSE150细胞分为空白对照组(未转染)、pcDNA3.1组和pcDNA3.1-FHIT组(转染相应载体),qRT-PCR检测细胞中FHIT mRNA表达水平,MTT法检测细胞增殖,Annexin V-FITC/PI双染法检测细胞凋亡,Western blot法检测细胞中NF-κB亚单位p65(NF-κBp65)、活化型Caspase-3(Cleaved Caspase-3)蛋白表达水平。结果:与癌旁正常组织比较,食管鳞癌组织中FHIT mRNA表达水平降低(P<0.05)。与空白对照组和pcDNA3.1组比较,pcDNA3.1-FHIT组KYSE150细胞中FHIT mRNA和蛋白表达水平升高,增殖活性降低,凋亡率和Cleaved Caspase-3蛋白表达水平升高,NF-κBp65蛋白表达水平降低(P<0.05)。结论:过表达FHIT可抑制食管鳞癌细胞增殖,诱导其凋亡,作用机制可能与抑制NF-κB信号通路的激活有关。Aim:To study the effects of fragile histidine triad(FHIT)overexpression on proliferation and apoptosis of esophageal squamous cell carcinoma cells and its possible mechanism.Methods:Real-time quantitative PCR(qRT-PCR)was used to detect the expression of FHIT mRNA in 59 cases of esophageal squamous cell carcinoma tissue and corresponding adjacent normal tissue.KYSE150 cells were divided into blank group,pcDNA3.1 group and pcDNA3.1-FHIT group.The expression of FHIT mRNA in the 3 groups was detected by qRT-PCR,cell proliferation was detected by MTT assay,apoptosis was detected by Annexin V-FITC/PI staining and the expressions of NF-κB subunit p65(NF-κBp65)and Cleaved Caspase-3 were detected by Western blot.Results:Compared with adjacent normal tissue,the expression of FHIT mRNA in esophageal squamous cell carcinoma tissue was significantly decreased(P<0.05).Compared with the blank group and pcDNA3.1 group,the expression of FHIT mRNA of pcDNA3.1-FHIT group was significantly increased,the survival rate decreased,apoptosis rate and Cleaved Caspase-3 protein expression increased,and NF-κBp65 protein expression decreased(P<0.05).Conclusion:Overexpression of FHIT could inhibit the proliferation of esophageal squamous cell carcinoma cells and induce apoptosis,which may be related to the inhibition of NF-κB signaling pathway.
