摘 要:目的:参附强心丸对2型心肾综合征模型大鼠肾脏的保护作用及HIF-VEGF-Notch信号通路的影响。方法:采用冠状动脉左前降支结扎联合肾切除的方法构建2型心肾综合征大鼠模型,实验分为4组:假手术对照组、心肾综合征模型组、参附强心丸治疗组、卡托普利组。治疗3周后,检测各组大鼠肾脏功能相关指标,如血尿素氮(BUN)、血肌酐(Scr)、尿肌酐(Ucr)以及尿硫酸吲哚酚(IS)的表达水平,HE染色观察肾脏细胞形态变化,Masson染色检测大鼠肾脏胶原含量以及纤维化程度,采用qRT-PCR以及Western blot对肾脏组织中HIF、VEGF以及Notch1的mRNA表达以及蛋白水平进行检测。结果:与假手术对照组相比,心肾综合征模型组大鼠肾脏功能相关指标BUN、Scr水平显著上升(P<0.05),Ucr、IS水平显著降低(P<0.05),参附强心丸治疗组大鼠肾脏功能指标BUN、Scr水平显著低于心肾综合征模型组(P<0.05),Ucr、IS水平显著升高(P<0.05)。参附强心丸组的HIF、VEGF以及Notch1的mRNA以及蛋白表达显著高于心肾综合征模型组。结论:参附强心丸对2型心肾综合征模型大鼠肾脏具有保护作用,且该作用可能与HIF-VEGF-Notch信号通路密切相关。Objective:To investigate the protective effect of Shen Fu QiangXin Pill on kidney and the influence of HIF-VEGF-Notch signaling pathway in rats with type 2 cardiorenal syndrome.Methods:The left anterior descending coronary artery ligation combined with partial nephrectomy was used to construct theα-type cardiorenal syndreme model rates.Then the model rats were randomly divided into four groups:sham operation control group,model group,Shen Fu Qiang Xin Pill treatment group and captopril group.Three weeks after treatment,related parameters of each rat renal function,such as blood urea nitrogen(BUN),serum creatinine(Scr),urine creatinine(Ucr)and uric acid indole phenol(IS)expression level were tested.The HE staining was used to observe the changes of renal cell morphology.The masson staining was employed to detect the contents of collagen in the rat kidney as well as the degree of fibrosis.The HIF,VEGF and Notch1 mRNA expression and protein levels in kidney tissue were detected independently by qRT-PCR and Western blot.Results:Compared with the sham operation control group,the renal function related indicators BUN,Scr significantly increased in the kidney syndrome model group(P<0.05),Ucr and IS significantly reduced(P<0.05).Renal function indicators BUN,Scr of group treated with Shen Fu Qiang Xin Pill was significantly lower than the model group(P<0.05),Ucr and IS significantly increased(P<0.05).mRNA and protein expressions of HIF,VEGF and Notch1 in Shen Fu Qiang Xin Pills group were significantly higher than those in the model group of cardiorenal syndrome.Conclusion:Shen Fu Qiang Xin Pill has a protective effect on the kidney of rats with type 2 cardiorenal syndrome,and this effect may be closely related to HIF-VEGF-Notch signaling pathway.
