摘 要:癌相关成纤维细胞(CAF)是肿瘤微环境中最丰富的细胞群,在肿瘤进展中起着至关重要的作用。CAF在肿瘤微环境中能分泌一些趋化因子,如IL-6、Chi3L1、SDF-1、CXCL12,这些因子通过旁分泌作用促血管生成、肿瘤侵袭及转移。这些因子不仅影响肿瘤免疫,还在肿瘤微环境中协助肿瘤细胞进行免疫逃逸,从而促进肿瘤进展和免疫耐受等。近年来,免疫检查点抑制剂在部分肿瘤晚期患者中应用,取得了较好的临床效果,部分患者得以长期存活。因此,靶向肿瘤微环境中特定的分泌因子与免疫检查点抑制剂联合应用,成为了研究的热点。本文综述了CAF的分泌组学及生物学功能的研究进展,并阐述其临床应用前景和面临的现实挑战。Cancer-associated fibroblasts(CAF)are the most abundant cells in tumor microenvironment and play a crucial role in tumor progression.CAF can secrete some chemokines in tumor microenvironment,such as IL-6,Chi3L1,SDF-1 and CXCL12,which promote angiogenesis,tumor invasion and metastasis through paracrine.These factors not only affect tumor immunity,but also assist tumor cells in immune escape in tumor microenvironment,thus promoting tumor progression and immune tolerance.In recent years,immunocheckpoint inhibitors have been used in some patients with advanced tumor,and achieved good clinical results,and some patients survived for a long time.Therefore,targeting specific secretory factors in tumor microenvironment and the combined application of immunocheckpoint inhibitors has become a research hotspot.In this paper,the research progress of secretory omics and biological functions of CAF are researched,and the clinical application prospect and realistic challenges are expounded.
