摘 要:目的:探究丙泊酚抑制wnt/β-catenin通路对宫颈癌HELA细胞生长和运动能力的作用机制。方法:体外培养人宫颈癌HELA细胞,分别使用0、2.5、5、10μg/ml的丙泊酚预处理后使用CCK8检测细胞增殖情况;使用流式细胞术检测细胞凋亡情况;使用Transwell检测细胞运动能力;使用Western blot检测Ki67、PCNA、Caspase-3、Caspase-9、MMP-9、VEGF表达情况;选取成年雌性Wistar大鼠60只,采用皮下注射宫颈癌HELA细胞悬液,制成宫颈癌移植瘤大鼠动物模型,腹腔注射低剂量(10 mg/kg)、中剂量(20 mg/kg)和高剂量(50 mg/kg)的丙泊酚,30 d后检测肿瘤重量并使用免疫组化检测宫颈癌组织Ki67和VEGF表达情况。结果:相比空白组,丙泊酚预处理组宫颈癌HELA细胞增殖能力、细胞凋亡率、Caspase-3蛋白表达水平、Caspase-9蛋白表达水平显著提高,且高剂量组显著高于中剂量组,中剂量组显著高于低剂量组,差异有统计学意义(P<0.05);细胞迁移能力、Ki67、PCNA、MMP-9、VEGF、wnt1、β-catenin、c-Myc蛋白表达水平、大鼠体内宫颈癌肿瘤质量、Ki67和VEGF表达阳性率显著降低,且高剂量组显著低于中剂量组,中剂量组显著低于低剂量组,差异有统计学意义(P<0.05)。结论:丙泊酚可以通过抑制wnt/β-catenin通路实现抑制宫颈癌HELA细胞生长和运动能力,且在一定范围内呈浓度依赖性。Objective:To explore the mechanism of propofol inhibition of wnt/β-catenin pathway on the growth and motor ability of cervical cancer HELA cells.Methods:Human cervical cancer HELA cells were cultured in vitro,and CCK8 was used to detect cell proliferation after preprocessing with propionol at 0,2.5,5 and 10μg/ml respectively.Flow cytometry was used to detect apoptosis.Transwell was used to detect cell motor ability.Western blot was used to detect the expression levels of Ki67,PCNA,Caspase-3,Caspase-9,MMP-9 and VEGF.60 adult female Wistar rats were selected and given subcutaneous injection of cervical cancer HELA cell suspension to prepare rat animal model of cervical cancer xenografts,and they were given intraperitoneal injection of low dose(10 mg/kg),middle dose(20 mg/kg)and high dose(50 mg/kg)of propofol.After 30 d,the tumor weight was detected,and immunohistochemistry was used to detect the expression levels of Ki67 and VEGF in cervical cancer tissues.Results:Compared with blank group,the proliferation ability of cervical cancer HELA cells,apoptosis rate and protein expression levels of Caspase-3 and Caspase-9 were significantly increased in propofol preprocessing groups,and the above indexes in high dose group were significantly higher than those in middle dose group,and the indexes in middle dose group were significantly higher than those in low dose group(P<0.05).Cell migration ability,protein expression levels of Ki67,PCNA,MMP-9,VEGF,wnt1,β-catenin and c-Myc,cervical cancer tumor quality,positive expression rates of Ki67 and VEGF in rats were significantly decreased,and the above indexes in high dose group were significantly lower than those in middle dose group,and the indexes in middle dose group were significantly lower than those in low dose group(P<0.05).Conclusion:Propofol can inhibit the growth and motor ability of cervical cancer HELA cells by inhibiting the wnt/β-catenin pathway,and it is concentration-dependent within a certain range.
