摘  要:经过表面消毒、研磨和稀释涂布,从海南大羽藓(Thuidium cymbifolium)叶片中分离得到一株对几种常见水果和作物病害的病原菌有较强抑制作用的内生细菌,通过形态观察及分子分析鉴定该菌为绿芽孢杆菌,编号为TC01。用比浊法测定TC01的生长曲线,发现其生长周期比普通细菌更长,2~6 d为稳定期;在此基础上,进行平板对峙实验和发酵液抑菌实验,对峙实验结果显示TC01对白绢病、香蕉枯萎病、立枯丝核病、芒果炭疽病等4种病原菌有不同程度的抑制作用,最大抑菌率分别是66.9%、59.7%、60.3%和83.0%;发酵液抑菌实验结果显示TC01对蔓枯病、稻瘟病、白绢病、香蕉枯萎病、立枯丝核病、芒果炭疽病等6种病原菌都有抑制作用,其中对白绢病病原菌和立枯丝核病病原菌的抑制效果最好,最大抑菌率分别高达92.8%和92.6%。内生细菌TC01具有较好的生防潜力,有深入研究和开发的价值。We isolated an endophytic bacterium with strong antifungal effect on diseases causal fungi of several common fruits and specific crops from the leaves of Thuidium cymbifolium in Hainan after surface disinfection,grinding,and dilution coating.Moreover,we identified the strain as Viridibacillus and named it as TC01 through morphological observation and molecular identification.Furthermore,we determined the growth curve of TC01 by a turbidimetric method,illustrating that the growth cycle of TC01 was longer than that of other ordinary bacteria and the growth stable period was 2-6 d after inoculation.Based on that,we performed the plate confrontation experiments and the fermentation broth antifungal experiments.The plate confrontation experiments show that TC01 presents different levels of antifungal activity against 4 pathogenic fungi,including Sclerotium rolfsii Sacc,Fusarium oxysporum,Rhizoctonia solani,and Colletotrichum gloeosporioides,and the maximum antifungal rates are 66.9%,59.7%,60.3%,and 83.0%,respectively.While the fermentation broth antifungal experiments show that TC01 presents antifungal activity against 6 pathogenic fungi,including Mycosphaerella melonis,Magnaporthe grisea,Sclerotium rolfsii Sacc,Fusarium oxysporum,Rhizoctonia solani,and Colletotrichum gloeosporioides.Of which,TC01 shows the best inhibitory effect against the pathogenic fungi of Sclerotium rolfsii Sacc and Rhizoctonia solani,and the maximum antifungal rates are 92.8%and 92.6%,respectively.In conclusion,TC01 has good biocontrol potential and has the value of further research and development.
