摘  要:目的:观察不同浓度地黄梓醇对木瓜蛋白酶诱导的早期膝骨关节炎大鼠膝关节滑膜组织中IL-1β、Galectin-3、S100A12蛋白表达的影响,探讨地黄梓醇对早期膝骨关节炎的作用机制。方法:40只3~8周龄成年雄性Wistar大鼠随机分成4组:正常对照组、模型对照组、地黄梓醇低、高剂量组,每组10只。正常对照组大鼠左膝关节腔内注射100μl生理盐水,其余3组大鼠左膝关节腔内注射等量4%木瓜蛋白酶复制早期膝骨关节炎大鼠模型。造模第10天随机抽取正常对照组大鼠4只、其余3组共4只大鼠,活体麻醉下进行左膝关节三维CT拍摄验证模型制作。各治疗组灌胃给予相应药物,正常对照组、模型对照组给予等体积生理盐水。28 d后,各组大鼠取左膝关节滑膜组织,免疫组化法检测各组大鼠滑膜组织中IL-1β、Galectin-3、S100A12蛋白水平。结果:三维CT结果显示造模的3组与正常对照组大鼠左膝关节间隙存在统计学差异(P<0.05)。模型对照组大鼠滑膜中S100A12、IL-1β、Galectin-3水平较正常对照组明显升高(P<0.01);地黄梓醇低剂量组大鼠滑膜中S100A12、IL-1β、Galectin-3水平较模型对照组有所降低(P<0.05);地黄梓醇高剂量组大鼠滑膜中S100A12、IL-1β、Galectin-3水平含量较模型对照组明显降低(P<0.01)。结论:不同浓度地黄梓醇均可降低膝骨关节炎大鼠滑膜组织中IL-1β、Galectin-3、S100A12蛋白水平,且地黄梓醇高剂量组较低剂量组效果更为明显,地黄梓醇可通过抑制炎症反应延缓膝骨关节炎病情进展。Objective:To observe effects of different concentrations of Rehmannia Catalpol on levels of IL-1β,Galectin-3 and S100A12 in synovial tissues of knee joint of rats with early knee osteoarthritis induced by papain,and to explore mechanism of Rehmannia Catalpol in treatment of early knee osteoarthritis.Methods:40 male Wistar rats aged 3-8 weeks were randomly divided into 4 groups of normal control group,model control group,low and high dose of Rehmannia Catalpol groups,10 in each group.In normal control group,100μl of normal saline was injected into left knee joint.The other three groups of rats were injected with same amount of 4%papain in left knee joint to replicate rat model of knee osteoarthritis.On the 10th day after modeled,4 rats in normal control group and 4 rats in other three groups were randomly selected.Three-dimensional CT imaging verification model of left knee joints were performed under anesthesia.Treatment groups were given corresponding drugs orrally,and normal control group and model control group were given equal volumes of normal saline.After 28 d,synovial tissues of left knee joints were taken from each group.Levels of S100A12 IL-1βand Galectin-3 in synovial tissue of each group were detected by immunohistochemistry.Results:Results of three-dimensional CT showed that there were statistical differences between three model groups and normal control group of left knee joints(P<0.05).Levels of S100A12,IL-1βand Galectin-3 in synovium of model control group were significantly increased than those in normal control group(P<0.01).Levels of S100A12,IL-1βand Galectin-3 in synovium of low dose group of rehmannia were lower than those in model control group(P<0.05).Levels of S100A12,IL-1βand Galectin-3 in synovium of high dose group of rehmannia were significantly lower than those in model control group(P<0.01).Conclusion:Different concentrations of rehmannia could reduce levels of IL-1β,Galectin-3 and S100A12 in synovial tissue of rats with knee osteoarthritis,and high dose of resveratrol was more
