摘　　要：目的:探究lncRNA UCA1靶向miR-145对卵巢癌细胞增殖和迁移的作用。方法:RT-qPCR检测卵巢癌细胞和正常卵巢上皮细胞UCA1和miR-145表达;在线数据库分析卵巢癌中UCA1表达;过表达和沉默UCA1,RT-qPCR检测miR-145表达;荧光素酶报告实验确定UCA1与miR-145的靶向关系;CCK8和Transwell实验检测细胞活性和迁移能力。结果:根据预实验结果选择SK-OV-3细胞进行实验。过表达UCA1可明显抑制miR-145表达(P<0.01),沉默UCA1可显著促进miR-145表达(P<0.01),减弱miR-145 inhibitor对miR-145表达的抑制作用(P<0.05);荧光素酶报告实验证实UCA1可靶向结合miR-145(P<0.05);细胞生物学实验证实沉默UCA1可显著抑制卵巢癌细胞增殖及迁移(P<0.05);下调UCA1显著抑制增殖和迁移相关蛋白PCNA、MMP-15表达。结论:沉默UCA1可靶向miR-145调节卵巢癌细胞的增殖和迁移能力。Objective:To investigate effects of lncRNA UCA1 on ovarian cancer cells via targeting miR-145.Methods:UCA1 and miR-145 levels of ovarian cancer cells and normal ovarian epithelial cells were measured by RT-qPCR.Expression of UCA1 in ovarian cancer was analyzed by online database.After silencing or over-expression UCA1,level of miR-145 was detected by RT-qPCR.Luciferase reporter assay was performed to determine relationship of UCA1 and miR-145.Cell proliferation and migration were detected by CCK8 and Transwell.Results:SK-OV-3 cell was selected for research study according to results of pre-experiment.Overexpression of UCA1 inhibited expression of miR-145 significantly(P<0.01),silencing UCA1 could significantly promote expression of miR-145(P<0.01),and attenuate inhibitory effect of miR-145 inhibitor on miR-145(P<0.05).Luciferase reporting experiments confirmed that UCA1 could target and bind to miR-145(P<0.05).Cell biology experiments confirmed that silencing UCA1 could significantly inhibit proliferation and migration of ovarian cancer cells(P<0.05).Down-regulation of UCA1 significantly inhibits expression of PCNA and MMP-15 proteins related to proliferation and migration.Conclusion:Down-regulation UCA1 can regulate proliferation and migration of ovarian cancer cells by targeting miR-145.
