摘 要:目的建立甘肃省地方习用药材椒目的质量标准。方法采用性状鉴别、显微鉴别及薄层色谱法建立椒目的定性鉴别方法,高效液相色谱法建立椒目中α-亚麻酸的含量测定方法,按《中国药典》(2015年)附录方法测定椒目中水分、总灰分、酸不溶性灰分、水溶性浸出物及醇溶性浸出物含量。结果优化的薄层色谱条件,可以特征性地实现11批椒目药材的定性鉴别,且11批样品均显示有α-亚麻酸的斑点。α-亚麻酸在0.011~1.344 mg/mL内呈良好的线性关系(r=0.9991),加样回收率为93.57%~110.88%,RSD=0.43%~2.93%;水分、总灰分、酸不溶性灰分、水溶性浸出物、醇溶性浸出物及α-亚麻酸含量分别为5.86%~8.74%、5.28%~6.65%、0.28%~1.22%、6.15%~9.45%、12.79%~25.48%、1.61%~4.42%。结论建立的薄层色谱法可以实现甘肃省地方习用药材椒目的定性鉴别,高效液相色谱法简便、可行,可用于椒目中α-亚麻酸的含量测定,为建立椒目质量标准提供依据。Objective To establish the quality standard of Zanthoxylum bungeanum Maxim.,which,also named Jiaomu,is one of the common medical herbs from Gansu Province.Methods Establishing a qualitative identification method of Jiaomu by microscopic identification,morphological identification and thin layer chromatography(TLC).Establishing a method to determine theα-linolenic acid by high performance liquid chromatography(HPLC).Meanwhile,the contents of moisture,total ash,acid insoluble ash,aqueous extracts and alcohol extracts were determined by the appendix of the Chinese Pharmacopoeia(2015 edition).Results The optimized TLC could characteristically realize the qualitative identification of 11 batches of Jiaomu and 11 batches of samples,all showingα-linolenic acid spots.The average content of theα-linolenic acid showed good linearity at the range of 0.011~1.344 mg/mL(r=0.9991),the recovery was 93.57%~110.88%and RSD was 0.43%~2.93%.The contents of moisture,total ash,acid insoluble ash,aqueous extracts,alcohol extracts andα-linolenic acid were 5.86%~8.74%,5.28%~6.65%,0.28%~1.22%,6.15%~9.45%,12.79%~25.48%,1.61%~4.42%,respectively.Conclusion The established TLC method can be used for a qualitative identification of the medicinal herb Jiaomu in Gansu Province.The HPLC method is simple and feasible,can be used to determine the content ofα-linolenic acid in Jiaomu.The above study can provide a basis for the establishment of quality standards of Jiaomu.
