摘　　要：目的研究垂体肿瘤转化基因-1(PTTG1)在急性髓系白血病(AML)中表达水平及其临床意义。方法收集初治和复发的AML患者骨髓标本,对照组收集非恶性血液肿瘤患者骨髓标本,荧光定量PCR方法检测PTTG1基因的表达水平,并分析其在不同临床特征、FAB分型和疾病分层的AML患者间的表达差异;根据AML患者外周血血细胞进行分类计数和骨髓幼稚细胞比例分组,分析不同组别间PTTG1的表达差异。结果PTTG1基因在AML患者骨髓细胞中表达水平明显高于对照组(P<0.05),初治AML和复发AML之间表达差异不具有统计学意义;不同FAB诊断分型中,M2型AML中PTTG1的表达水平最高,M1型中表达最低,但是差异无统计学意义(P>0.05);根据疾病危险分层分析,低危组中PTTG1的表达水平最高,高危组中PTTG1的表达水平最低,但组间差异无统计学意义(P>0.05);重度贫血和中度贫血AML患者中PTTG1的表达水平明显低于轻度贫血和正常AML患者,差异有统计学意义(P<0.05);根据外周血白细胞和血小板计数以及骨髓幼稚细胞比例设定的不同组别中,PTTG1的表达差异无统计学意义(P>0.05)。结论PTTG1在AML患者骨髓细胞中表达水平明显高于对照组,其表达水平可能与患者的贫血程度有关。Objective To study the expression level of pituitary tumor transforming gene-1(PTTG1)in acute myeloid leukemia(AML)and and its clinical significance.Methods Bone marrow samples were collected from newly diagnosed and relapsed AML patients and non malignant blood tumor patients.The expression level of PTTG1 gene was detected by qPCR.According to the clinical characteristics,FAB classification,disease stratification,peripheral blood cell counts and the proportion of immature bone marrow cells in AML patients,the expression difference of PTTG1 among different groups was analyzed.Results The expression level of PTTG1 gene in bone marrow cells of AML patients was significantly higher than that of control group(P<0.05),and there was no significant difference between the expression of PTTG1 between newly diagnosed AML and relapsed AML;The expression level of PTTG1 in M2 AML was the highest and that in M1 type was the lowest,but the difference was not statistically significant(P>0.05).According to the disease risk stratification analysis,the expression level of PTTG1 in low-risk group was higher and that in high-risk group was the lowest,but the difference was not statistically significant(P>0.05);The expression level of PTTG1 in severe anemia and moderate anemia AML patients was significantly lower than that in mild anemia and normal AML patients(P<0.05);There was no significant difference in the expression of PTTG1 in different groups according to the peripheral blood leukocyte and platelet count and the proportion of immature bone marrow cells(P>0.05).Conclusion The expression level of PTTG1 in AML patients was significantly higher than that of the control group,and the expression level of PTTG1 may be related to the degree of anemia in patients with AML.
