摘　　要：目的探讨新型生物标志物肿瘤坏死因子α诱导蛋白8样2(tumor necrosis factor-αinduced protein 8-like 2,TIPE2)在透明细胞肾癌患者肿瘤、癌旁组织及血清中的表达及其临床意义。方法选取2018年1~12月,山东省千佛山医院和上海芯超生物有限公司透明细胞肾细胞癌肿瘤及癌旁石蜡组织切片共65例,免疫组化方法检测TIPE2表达。选取同期来自山东省千佛山医院65名健康体检者作为对照组,用ELISA方法,检测透明细胞肾细胞癌患者及正常人群外周血清中TIPE2蛋白表达,并分析TIPE2表达水平在透明细胞肾癌中的临床意义。结果免疫组化结果显示,TIPE2蛋白在透明细胞肾细胞癌肿瘤组织特异性高表达,在癌旁组织中弱表达(P<0.01)。TIPE2蛋白水平在透明细胞肾癌患者组血清中高于正常对照组(P<0.01)。结论TIPE2在透明细胞肾细胞癌肿瘤及血清中的特异性高表达,有望成为新型生物标志物,有助于透明细胞肾细胞癌的早期诊断。Objective To explore the expression and clinical significance of tumor necrosis factor-αinduced protein 8-like 2(TIPE2)in tumor,adjacent tissues and serum of patients with in clear cell renal cell carcinoma(ccRCC).Methods From January to December 2018,65 paraffin tissue samples of ccRCC from Shandong provincial Qianfoshan hospital and Shanghai outdo biotech co,ltd were selected.The levels of TIPE2 in tumor and adjacent tissues were detected by immunohistochemistry.65 healthy people from Shandong provincial Qianfoshan hospital were involved as control group.The expression of TIPE2 protein in serum of ccRCC patients and control group was tested by ELISA method.In the meanwhile,the clinical significance of TIPE2 expression was analyzed in the diagnosis of clear cell renal cell carcinoma.Results TIPE2 protein was specific over-expressed in ccRCC tumor tissues,but weakly expressed in adjacent tissues(P<0.01).The level of TIPE2 protein in serum of patients with ccRCC was significantly higher than that of normal control group too(P<0.01).Conclusion As a new potential biomarker,TIPE2 has high specific expression in tumor tissue and serum of patients with ccRCC,which will be helpful for early diagnosis of clear cell renal cell carcinoma.
