纵条纹炭角菌子实体的急性毒性试验及改善睡眠作用研究

摘　　要：为确定纵条纹炭角菌子实体的安全性及对改善睡眠的效应,通过急性毒性试验评价子实体安全性,并确定最大给药量;通过小鼠戊巴比妥钠阈剂量(阈下剂量)催眠试验,探究纵条纹炭角菌子实体对小鼠睡眠的效应。急性毒性试验显示纵条纹炭角菌子实体的最大给药量为2 000 mg/kg,小鼠的生理状况没有明显改变且在组织病理学分析中不存在毒性表现;戊巴比妥钠诱导的睡眠实验显示该菌子实体在高剂量(2 000 mg/kg)时可以显著增加入睡小鼠数量,延长小鼠睡眠时间并减少小鼠睡眠潜伏期(P<0.01)。表明其可以用于失眠症的治疗。To investigate the safety of Xylaria striata and its effect on mice sleep improvement. The safety of the fruiting body of X. striata was evaluated by acute toxicity tests, and the maximum dose of X. striata was determined. The mouse pentobarbital sodium threshold dose(threshold sub-dose) hypnosis test was used to evaluate the effect of sleep in mice. Results: The acute toxicity test of fruiting body of X. striata showed that the maximum dose a mouse can take exceeds 2 000 mg/kg. The physiological condition of the mice did not change significantly and there were no toxic symptoms in histopathological analysis. Pentobarbital-induced sleep experiments showed that the fruiting body of X. striata increased significantly the number of mice, prolonged the sleep time of mice and reduced the sleep latency of mice(P<0.01) in high dose of 2 000 mg/kg. Conclusion: Under the maximum dose of 2 000 mg/kg, mice showed no signs of toxicity, so the fruiting body of X. striata is highly safe for oral administration. And it has the obvious improvement sleep activity, indicating its possible therapeutic use for insomnia.
