摘  要:目的探讨姜黄素(Curcumin,Cur)对人结直肠癌HT-29细胞增殖、凋亡的影响,分析其可能存在的作用机制。方法体外培养人结直肠癌HT-29细胞,分为对照组与干预组,干预组再分为3个亚组并进行不同浓度Cur(20、40、80μmol/L)处理,Cell Counting Kit-8(CCK-8)法检测细胞增殖情况,流式细胞法检测细胞周期分布情况,实时逆转录-聚合酶链反应(real-time RT PCR,RT-PCR)检测各组细胞周期相关基因表达量,流式细胞法检测细胞凋亡率,Western blot检测细胞凋亡、Wnt/β-catenin信号通路相关蛋白水平。结果干预组12、24、48 h细胞增殖抑制率均明显高于对照组(P<0.05),随着Cur浓度增加,细胞增殖抑制率逐渐升高(P<0.05)。细胞周期检测结果显示,与对照组相比,干预组G0/G1期细胞百分比升高(P<0.05),S期细胞百分比降低(P<0.05)。RT-PCR结果显示,干预组CyclinD1、Survivin、C-myc表达量随着Cur浓度增加而逐渐降低(P<0.05)。细胞凋亡检测结果显示,随着Cur浓度增加,干预组活细胞百分比、Bcl-2逐渐降低(P<0.05),而早期凋亡细胞百分比、晚期细胞凋亡百分比、Bax水平、cleaved-Caspase-3水平逐渐升高(P<0.05)。Western blot检测结果显示,随着Cur浓度增加,干预组p-β-catenin、Wnt3a水平逐渐降低(P<0.05),而β-catenin水平变化不明显(P>0.05)。结论Cur可以抑制人结直肠癌HT-29细胞增殖、促进细胞凋亡,可能与调节细胞周期相关基因表达量、凋亡蛋白水平及Wnt/β-catenin信号通路活化有关。Objective To explore the effects of curcumin(Cur)on proliferation and apoptosis of human colorectal cancer HT-29 cells,and to analyze the possible action mechanisms.Methods Human colorectal cancer HT-29 cells were cultured in vitro.They were divided into control group and intervention group.The intervention group was further divided into three subgroups,which were treated with different concentration of Cur(20,40,80μmol/L).The cell proliferation was detected by Cell Counting Kit-8(CCK-8).The distribution of cell cycle was detected by flow cytometry.The expression levels of cell cycle related genes were detected by real-time PCR(RT-PCR).The apoptosis rate was detected by flow cytometry.The levels of apoptosis and Wnt/β-catenin signaling pathway-related proteins were detected by Western blot.Results At 12 h,24 h and 48 h,inhibition rate of cell proliferation in intervention group was significantly higher than that in control group(P<0.05).With increase of Cur concentration,inhibition rate of cell proliferation was increased gradually(P<0.05).The test results of cell cycle showed that compared with control group,percentage of cells in G0/G1 phase was increased in intervention group(P<0.05),while percentage of cells in S phase was decreased(P<0.05).RT-PCR results showed that expression levels of CyclinD1,survivin and C-myc in intervention group were decreased gradually with increase of Cur concentration(P<0.05).The apoptosis test results showed that with increase of Cur concentration,percentage of viable cells and Bcl-2 in intervention group were decreased gradually(P<0.05),while percentages of early apoptotic cells and late apoptosis,levels of Bax and cleaved-Caspase-3 were increased gradually(P<0.05).Western blot results showed that with increase of Cur concentration,levels of p-β-catenin and Wnt3a in intervention group were decreased gradually(P<0.05),while changes inβ-catenin level were not significant(P>0.05).Conclusion Cur can inhibit proliferation of human colorectal cancer HT-29 cells,and promote their ap
