摘 要:目的:分析并鉴定STGC3基因转录调控元件,探讨鼻咽癌STGC3基因的转录调控分子机制。方法:运用生物信息学,预测并分析STGC3基因核心启动子区的转录因子结合位点;将相关转录因子结合位点,制备3种探针即生物素标记野生型、突变型及未标记野生型;提取CNE2细胞核蛋白,利用电泳迁移率变动分析(EMSA)及染色质免疫共沉淀(ChIP)分析,体内外鉴定转录因子结合位点。结果:在STGC3基因核心启动子区存在21个转录因子结合位点,其中9个为Sp1转录因子结合位点;进一步严格限定参数,STGC3基因核心启动子区域含有转录因子结合位点5个,其中Sp1转录因子结合位点2个;EMSA和ChIP实验结果显示,STGC3基因中存在转录因子Sp1特异性结合位点,从而鉴定出STGC3基因转录调控元件。结论:STGC3基因核心启动子区含有Sp1特异性结合位点,为该基因的转录调控元件。Objective:The transcription regulatory elements of STGC3 gene were analyzed and identified to explore the molecular mechanism of transcription regulation of STGC3 gene in nasopharyngeal carcinoma(NPC).Methods:Transcriptional factor binding sites in the core promoter region of the STGC3 gene was predicted and analyzed by bioinformatics.Based on the sequence of related transcriptional factor binding sites,three kinds of probes including namely biotin labeled wild type,mutant type and unlabeled wild type,were prepared.CNE2 nuclear protein was extracted,and transcription factor binding sites were identified in vitro and in vivo by electrophoretic mobility change(EMSA)and chromatin immunoprecipitation(ChIP)analysis.Results:The result showed that there were 21 transcription factor binding sites in core promoter region of the STGC3 gene,9 of which were Sp1 transcription factor binding sites.If the parameters were further strictly limited,there were 5 transcription factor binding sites in core promoter region of the STGC3 gene,2 of which were SP1 transcription factor binding sites.EMSA and ChIP experimental results showed that there was a specific binding site of transcription factor Sp1 in STGC3 gene.Conclusion:The core promoter of STGC3 gene contains Sp1 specific binding site,which is a transcription regulatory element of STGC3 gene.
