摘 要:随着土壤盐碱化面积的扩大,作为第四大主粮作物的马铃薯耐盐品种的选育和耐盐机理的研究尤为重要。本试验以六个马铃薯组培苗品种为材料,研究在不同NaCl浓度下生理特性的变化。结果表明,随着NaCl浓度升高,‘尤金885’的叶绿素A、B及类胡萝卜素含量降低幅度最小,丙二醛(MDA)含量增长最迅速,过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性最强。利用NCBI数据库进行比对,筛选到耐盐基因StDWF4,并对其进行生物信息学分析。结果表明,StDWF4基因编码1476个氨基酸,等电点为5.04,是一种含信号肽的亲水性、分泌型蛋白。二级结构分析显示,该蛋白含有α螺旋169处,占总二级结构的34.42%;延伸链104处,占21.18%;无规则卷曲218处,占44.40%。同源性比对及系统进化树分析表明,StDWF4基因编码的氨基酸序列与番茄cytochrome P45090B3耐盐基因亲缘关系为最近。本研究结果表明,‘尤金885’的耐盐性最强,为今后耐盐品种的选育提供良好材料;生物信息学分析表明,StDWF4基因具有耐盐基因的典型特征,为马铃薯耐盐品种的分子鉴定提供理论指导。With the increase of soil salinization area,the selection and breeding of salt-tolerant varieties and the study of salt-tolerant mechanism of potato are particularly important.In our study,tissue culture plants of six potato varieties were used to study the variations of physiological characteristics under different salt stress of sodium chloride(NaCl).The results showed that with the increase of NaCl concentrations,the content of chlorophyll A,chlorophyll B and carotenoids decreased in variety’Youjin885’less than those in other varieties,the content of malondialdehyde(MDA)increased in variety’Youjin885’more than that in other varieties,and the activities of catalase(CAT)and superoxide dismutase(SOD)were the strongest in variety’Youjin885’when compared with other varieties.Salt tolerance gene StDWF4 was found when using the NCBI BLAST,and then bioinformatics analysis was conducted.The results demonstrated that StDWF4 gene encoded 1476 amino acids,a hydrophilic and secretory protein containing signal peptides,and its isoelectric point was 5.04.Secondary structure analysis showed that the protein contained 169 alpha-helices,104 extended chains and 218 irregular curls,and accounting for 34.42%,21.18%and 44.40%of the total secondary structure,respectively.Homology comparison and phylogenetic tree analysis showed that the amino acid sequence encoded by the StDWF4 gene was closest to that of the salt tolerance gene CYP90 B3 in tomato.The results of this study indicated that the’Youjin885’was the best salt-tolerant variety and the gene StDWF4 had typical characteristic of salt tolerance genes,which play an important role for breeding and identification of salt-tolerant varieties.
