摘 要:为了探讨水龙骨目FLO/LFY同源基因的分子遗传与进化关系。本研究通过PCR技术获得水龙骨目(Polypodiales)13科14种蕨类植物FLO/LFY同源基因全长DNA序列。与肾蕨(Nephrolepis auriculata)和荚果蕨(Matteuccia struthiopteris)LFY同源基因DNA全长序列相结合,对水龙骨目FLO/LFY同源基因序列进行比较分析。结果表明:每个FLO/LFY同源基因DNA全长序列均含3外显子和2内含子,其第一内含子和第二内含子长短不同,差异明显。与其他蕨类植物中的FLO/LFY同源基因的同源性较高(72.13%~95.20%)。此外,用FLO/LFY同源基因的氨基酸序列、第一内含子序列和第二内含子序列构建蕨类植物的系统进化树。结果表明:由FLO/LFY同源基因第一内含子序列构建的系统进化树能正确反映水龙骨目科间的系统进化关系,与传统形态学的分类系统和分子系统学的分类系统较一致,更适合蕨类植物FLO/LFY同源基因系统进化树的构建。In order to explore the molecular genetic and evolutionary relationship of FLO/LFY homologous genes of Polypodiales.The full-length DNA sequences of FLO/LFY homologous genes were obtained from 14 ferns of13 families in Polypodiales by PCR.The sequence of FLO/LFY homologous gene in Polypodiales was compared and analyzed by combining the corresponding DNA sequence of LFY homologous gene in Nephrolepis auriculata and Matteuccia struthiopteris.The results showed that:each FLO/LFY homologous gene DNA fragment contains3 exons and 2 introns.The length of the intron 1 and the intron 2 were different,and the difference was obvious.The FLO/LFY homologous gene was highly homologous with other ferns(72.13%~95.20%).In addition,phylogenetic trees of ferns were constructed by amino acid sequence,intron 1 sequence and intron 2 sequence of FLO/LFY homologous gene.The results showed that:the phylogenetic tree constructed from the intron 1 sequence of FLO/LFY homologous gene can correctly reflect the phylogenetic relationship among Polypodiales,which is more consistent with the traditional morphological and molecular phylogenetic taxonomic systems,and is more suitable for the construction of phylogenetic tree of FLO/LFY homologous gene in ferns.
