摘 要:为寻找天然抗氧化剂,对翅茎白粉藤乙醇浸膏体内抗氧化活性进行研究并探讨其作用机制。将昆明种小鼠随机分为空白组、模型组、低剂量组、中剂量组和高剂量组,以灌胃给药的方式喂养15 d,末次灌胃后采用乙醇氧化损伤的方法造模后分离血清,摘取肝脏和肾脏,比较各组小鼠血清中MDA(丙二醛)含量、GSH(还原型谷胱甘肽)含量、SOD(超氧化物歧化酶)活力、GSH-Px(谷胱甘肽过氧化物酶)活力以及肝脏系数和肾脏系数。结果表明:与空白组比较,模型组小鼠血清中MDA含量显著高于空白组(p<0.01),而血清中GSH含量、SOD活力、GSH-Px活力均低于空白组,差异具有统计学意义(p<0.01),并且模型组肾脏系数和肝脏系数均显著下降(p<0.05);与模型组比较,低、中、高剂量组小鼠血清中MDA含量均显著降低(p<0.05),GSH含量、SOD活力、GSH-Px活力均显著升高(p<0.05),肾脏系数和肝脏系数无显著差异(p>0.05)。翅茎白粉藤乙醇浸膏具有明显的体内抗氧化活性,其作用机制可能与降低体内脂质过氧化产物、清除自由基和增强抗氧化酶活性有关。In order to select the best natural antioxidants,the antioxidative activity of ethanol extract from Cissus hexangularis in vivo and its mechanism were analyzed.Kunming mice were randomly divided into blank group,model group,low dose group,middle dose group and high dose group.Mice stomachs were perfused with ethanol extract from C.hexangularis for 15 days,the mice model was established by the method of ethanol oxidative injury after the last intragastric gavage,the serum was separated,and the liver and kidney were removed.The contents of MDA and GSH,the activities of GSH-Px and SOD in the serum and the liver coefficient and kidney coefficient of each group of mice were compared.The content of MDA in the model group was significantly higher than that in the blank group(p<0.01),while the content of GSH,the activity of SOD,and the activity of GSH-Px in the serum of the model group were significantly lower than those in the blank group(p<0.01),and the kidney and liver coefficients were significantly lower in the model group(p<0.05).The levels of MDA in the low,middle and high groups were significantly lower than those in the model group(p<0.05),and the content of GSH,the activity of SOD,the activity of GSH-Px in the serum of each group were significantly higher than those in the model group(p<0.05).In addition,the kidney and liver coefficients of the three dose groups are higher than those of the model group,but not significantly different(p>0.05).The ethanol extract from C.hexangularis had obviously antioxidative activity in vivo,and its mechanism may be related to reducing of the lipid peroxidation products,scavenging of free radicals and enhancing of the activity of antioxidase.
