摘　　要：目的观察长链非编码RNA(lncRNA)C00473在子宫内膜癌组织中的表达,及其通过调控微小RNA 15b(miR-15b)对子宫内膜癌细胞(Ishikawa)增殖能力的影响。方法qRT-PCR检测20例子宫内膜癌组织(研究组)与20例正常子宫内膜组织(正常组)中lncRNA-C00473的表达情况。在Ishikawa细胞中转染过表达质粒(pcDNA3.1-C00473)、siRNA-C00473以及空质粒(pcDNA3.1),分别为过表达组、抑制组及对照组(NC组)。CCK8法检测各组Ishikawa细胞增殖情况。qRT-PCR、western-blot检测各组中lncRNA-C00473、miR-15b及其靶基因细胞周期蛋白D1(cyclin D1)的表达情况。结果与正常组相比,研究组lncRNA-C00473的表达增高(P<0.05)。qRT-PCR显示Ishikawa细胞中lncRNA-C00473过表达与抑制效果显著(P<0.05);与NC组相比,过表达组细胞增殖能力升高,抑制组细胞增殖力降低,差异有统计学意义(均P<0.05);过表达组miR-15b水平低于抑制组,cyclin D1在mRNA和蛋白水平均高于抑制组,差异有统计学意义(P<0.05)。结论lncRNA-C00473与子宫内膜癌相关,其可能通过miR-15b/cyclin D1途径影响子宫内膜癌细胞增殖能力。Objective To observe the expression of long non-coding RNA(lncRNA)C00473 in endometrial carcinoma tissues and its effect on the proliferation ability of endometrial carcinoma cells(Ishikawa cells)by regulating microRNA 15b(miR-15b).Methods qRT-PCR was used to detect the expression of lncRNA-C00473 in 20 cases of endometrial carcinoma tissues(research group)and 20 cases of normal endometrial tissues(normal group).The over-expression plasmid(pcDNA3.1-C00473),siRNA-C00473,and empty plasmid(pcDNA3.1)were transfected into Ishikawa cells and divided into overexpression group,suppression group,and control group(NC group).CCK8 method was used to detect the proliferation of Ishikawa cells in each group.qRT-PCR and western-blot were used to detect the expression of lncRNA-C00473,miR-15b and its target gene cyclin D1(cyclin D1)in each group.Results Compared with the normal group,the expression of lncRNA-C00473 in the study group increased(P<0.05).qRT-PCR showed that lncRNA-C00473 overexpression and inhibitory effect were significant in Ishikawa cells(P<0.05).The cell proliferation ability of the expression group was increased,and the cell proliferation of the inhibition group was decreased,and the difference was statistically significant(P<0.05).The level of miR-15b in the overexpression group was lower than that in the inhibition group,and the levels of cyclin D1 in the overexpression group were higher than those in the mRNA and protein levels.In the inhibition group,the difference was statistically significant(P<0.05).Conclusion lncRNA-C00473 is associated with endometrial carcinoma,which may affect the proliferation ability of endometrial carcinoma cells through miR-15b/cyclin D1 pathway.
