摘　　要：目的检测涎腺腺样囊性癌中微小RNA-22-3p(miR-22-3p)的表达,探讨其临床意义及与血管内皮生长因子-C(VEGF-C)和微淋巴管密度(MLVD)的关系。方法选取我院2011年1月~2014年7月收治的61例涎腺腺样囊性癌患者为研究对象,留取肿瘤组织(观察组)和距肿物边缘>2 cm的正常涎腺组织(对照组)。应用实时荧光定量PCR法检测两组中miR-22-3p的表达,免疫组化方法检测观察组中VEGF-C和D2-40的表达,Western Blot法检测观察组中VEGF-C的表达。结果观察组中miR-22-3p的表达明显低于对照组,且观察组中miR-22-3p的表达在不同肿瘤直径、血管淋巴管累犯、有无远处转移、淋巴结转移和不同TNM分期中差异均有统计学意义(均P<0.05),而在不同性别、年龄、神经累犯中差异均无统计学意义(均P>0.05)。生存分析显示,miR-22-3p低表达患者的生存时间短。miR-22-3p与VEGF-C、MLVD呈负相关。结论miR-22-3p在涎腺腺样囊性癌中低表达,可能与病变的进展及淋巴管生成有关。miR-22-3p可能是涎腺腺样囊性癌独立的预后因子。Objective To detect the expression of microRNA-22-3p(miR-22-3p)in salivary adenoid cystic carcinoma,explore its clinical significance and pay attention to its relationship with vascular endothelial growth factor-C(VEGF-C)and microlymphatic vessel density(MLVD).Methods 61 patients with adenoid cystic carcinoma of salivary gland were selected as the study subjects.Tumor tissue(observation group)and normal salivary gland tissue(control group)were retained.Expression of miR-22-3p were detected by real-time fluorescence quantitative PCR in the two groups.VEGF-C and D2-40 were detected by immunohistochemical method in the observation group.VEGF-C were detected by Western Blot method in the observation group.Results The expression of miR-22-3p was significantly lower in the observation group than that in the control group.The expression of miR-22-3p was significantly different in tumor diameter,vascular and lymphatic vessel involvement,metastasis,lymph node metastasisand TNM stage,but there was not significantly different in gender,age and nerve involvement in the observation group.Survival analysis showed that patients of lower expression of miR-22-3p had shorter survival time.Negative correlation were found between miR-22-3p and VEGF-C,miR-22-3p and MLVD.Conclusion The expression of miR-22-3p is low in salivary adenoid cystic carcinoma,which is related to the progression.The low expression of miR-22-3p may be related to lymphangiogenesis.MiR-22-3p may be an independent prognostic factor for salivary adenoid cystic carcinoma.
