miR-211-5p和p27^Kip1在原发性胃癌组织中的表达水平与临床病理学特征的相关性研究

摘　　要：目的探究miR-211-5p和p27^Kip1在胃癌组织中的表达水平,以及其与患者临床病理学特征和预后的关系。方法收集2015年8月至2016年8月在开滦总医院林西医院接受手术治疗的60例胃癌患者的临床资料,采用实时定量聚合酶链反应和免疫组织化学法检测胃癌组织、癌旁组织(距离肿瘤边缘>2 cm)中miR-211-5p和p27^Kip1的表达水平。探讨miR-211-5p和p27^Kip1阳性率与患者的临床病理学特征和预后的相关性。采用Kendall′s tau-b等级相关系数分析miR-211-5p与p27^Kip1表达的相关性。结果与癌旁组织相比,胃癌组织中miR-211-5p的阳性率显著升高,而p27^Kip1的阳性率则显著降低。miR-211-5p和p27^Kip1的阳性率与患者的TNM分期、淋巴结转移和浸润深度有关(P<0.05),而与性别、年龄、肿瘤位置和大小不相关(P>0.05)。miR-211-5p与p27^Kip1在胃癌组织中的表达水平呈负相关(r=-0.455,P<0.031),miR-211-5p阳性表达者的3年生存率显著低于阴性表达者(log-rankχ^2=19.85,P<0.0001);而p27^Kip1阳性表达者的3年生存率则高于阴性表达者(log-rankχ^2=17.53,P<0.0001)。结论在胃癌组织中miR-211-5p呈高表达,p27^Kip1呈低表达,两者呈负相关,且其表达水平的异常提示胃癌患者的预后不良。Objective This paper is to explore the expression of miR-211-5p and p27^Kip1 in gastric cancer tissues and their correlation with clinicopathological features and prognosis.Methods The clinical data of 60 patients with gastric cancer who underwent surgery in Linxi Hospital,Kailuan General Hospital from August 2015 to August 2016 were collected.Real-time quantitative polymerase chain reaction and immunohistochemistry were used to detect the expression of miR-211-5p and p27^Kip1 in tumor tissues and paracancerous tissues(distance>2 cm from tumor tissues)of gastric cancer patients.Besides,the correlation between the positive rate of miR-211-5p,p27^Kip1 and the clinicopathological features and prognosis of gastric cancer patients were also analyzed.Kendall′s tau-b was used to analyze the correlation between the expression of miR-211-5p and p27^Kip1.Results Compared with paracancerous tissues of gastric cancer patients,the positive rate of miR-211-5p in tumor tissues was significantly increased,while the positive rate of p27^Kip1 was significantly decreased.The positive rates of miR-211-5p and p27^Kip1 were significantly correlated with the TNM stage,lymph node metastasis,and depth of invasion in tumor tissues(P<0.05),but not with gender,age,tumor location,and size(P>0.05).There was a negative correlation between the expression of miR-211-5p and p27^Kip1 in gastric cancer patients(r=-0.455,and P<0.031).The 3-year survival rate of gastric cancer patients with positive expression of miR-211-5p was significantly lower than that of patients with miR-211-5p negative expression(log-rankχ^2=19.85,and P<0.0001),while that of patients with positive expression of p27^Kip1 was higher than that of patients with negative expression(log-rankχ^2=17.53,and P<0.0001).Conclusions The expression of miR-211-5p was high while that of p27^Kip1 was low in gastric cancer tissues,and there was a negative correlation between the two.The abnormal expression of miR-211-5p and p27^Kip1 suggest that the prognosis of gastric cancer patients is
