摘  要:目的研究姜黄素通过调控miR-425表达水平对炎症性肠病(IBD)大鼠炎性反应的影响。方法选取30只成年雄性SD大鼠作为研究对象,随机分为对照组、IBD组和IBD+姜黄素组,每组10只。采用三硝基苯磺酸(TNBS)灌胃的方法构建IBD模型,对照组给予等量15%乙醇溶液灌胃。建模后,IBD+姜黄素组每日给予姜黄素(100 mg/kg)灌胃,IBD组每日给予蒸馏水(100 mg/kg)灌胃。连续灌胃7 d后,采集各组的静脉血,采用逆转录-聚合酶链反应(RT-PCR)检测miR-425表达水平,采用流式细胞术(FCM)检测辅助性T细胞17(Th17细胞)和调节性T细胞(Treg细胞)的百分比,采用酶联免疫吸附试验(ELISA)检测促炎因子白细胞介素-6(IL-6)、IL-17和抑炎因子转化生长因子-β(TGF-β)的表达水平。喂养期间观察各组的体质量、粪便性状和隐血情况,并比较各组疾病活动度指数(DAI)的差异。结果与对照组相比,IBD组静脉血中miR-425表达水平升高(P=0.002),Th17细胞百分比升高(P<0.001)、Treg细胞百分比降低(P=0.008),IL-6、IL-17表达水平升高(P<0.001)、TGF-β表达水平降低(P<0.001),DAI评分升高(P=0.004)。与IBD组相比,IBD+姜黄素组miR-425表达水平降低(P=0.002),Th17细胞百分比降低(P<0.001)、Treg细胞百分比升高(P=0.008),IL-6、IL-17表达水平降低(P=0.002,P<0.001)、TGF-β表达水平升高(P<0.001),DAI评分降低(P=0.004)。结论姜黄素可通过下调miR-425表达水平来减轻IBD大鼠的炎性反应和改善病情。Objective This paper intends to study the effect of curcumin on the inflammatory response of rats with inflammatory bowel disease(IBD)by regulating the expression of miR-425.Methods Thirty adult male SD rats were selected as the research objects and randomly divided into the control group,the IBD group,and the IBD+curcumin group,with 10 rats in each group.The IBD model was constructed by intragastric administration of trinitrobenzene sulfonic acid(TNBS),and the control group was given the same amount of ethanol by gavage.After modeling,the IBD+curcumin group was given curcumin(100 mg/kg)daily by gavage,and the IBD group was given distilled water(100 mg/kg)daily by gavage.After continuous intragastric gavage for 7 days,venous blood was collected from each group.The reverse transcription-polymerase chain reaction(RT-PCR)was used to detect the expression of miR-425,the flow cytometry(FCM)was used to detect the percentage of Th17 cells and Treg cells,and the enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of pro-inflammatory factors interleukin-6(IL-6),IL-17 and anti-inflammatory factor transforming growth factor-β(TGF-β).During the feeding period,the body weight,fecal traits,and occult blood of each group were observed,and the differences in disease activity index(DAI)of each group were compared.Results Compared with the control group,the expression of miR-425 in the venous blood of the IBD group increased(P=0.002),the percentage of Th17 cells increased(P<0.001),the percentage of Treg cells decreased(P=0.008),the expression of IL-6 and IL-17 increased(P<0.001),the expression of TGF-βdecreased(P<0.001),and the DAI score increased(P=0.004).Compared with the IBD group,the expression of miR-425 in the IBD+curcumin group decreased(P=0.002),the percentage of Th17 cells decreased(P<0.001),the percentage of Treg cells increased(P=0.008),the expression of IL-6,IL-17 decreased(P=0.002,P<0.001),the expression of TGF-βincreased(P<0.001),and the DAI score decreased(P=0.004).Conclusion Curcumin can r
