摘　　要：小窝蛋白-1(Cav-1)是一种支架蛋白,对恶性肿瘤的发生、发展起关键调控作用。在恶性肿瘤起始阶段,Cav-1可促进肿瘤血管新生并介导肿瘤干细胞无限增殖及自我更新;在恶性肿瘤转移阶段,Cav-1可参与肿瘤细胞的黏附运动、失巢凋亡和自噬形成;此外,Cav-1的表达可直接或间接促进肿瘤多药耐药,同时也能影响肿瘤细胞与间质微环境的相互作用。然而,不同肿瘤或同一肿瘤不同阶段的Cav-1表达具有显著差异性和两面性,因此Cav-1作为抑癌因子或致癌因子一直存在争议。深入研究Cav-1信号转导通路及其与肿瘤的关系,有望为诊断、治疗、预防相关肿瘤提供新途径。Caveolin-1(Cav-1)is a scaffold protein,which plays an important role in the occurrence and development of malignant tumors.Cav-1 not only promotes angiogenesis and mediates the unlimited proliferation and self-renewal ability of cancer stem cells during tumorigenesis stage,but also participates in the adhesion movement,anoikis and autophagy of tumor cells during metastatic progression.In addition,the expression of Cav-1 stimulates the multidrug resistance of cancer in either a direct or an indirect way,and affects the interaction between tumor cells and interstitial microenvironment.Nevertheless,the expressions of Cav-1 in different tumors or different stages within a single tumor has significant differences in its expressional levels,leading to the controversies of its dual role as oncogene or tumor suppressor.A thorough investigation of Cav-1 signaling pathway and its interaction with tumors will provide a novel approach for the diagnosis,treatment and prevention of the tumors.
