摘  要:目的探讨LYPD3在肺腺癌(LUAD)中的作用及预测临床预后的价值。方法检索癌症基因组图谱(TCGA)获得相关数据:535例LUAD组织标本及59例癌旁组织标本LYPD3基因表达及DNA甲基化表达情况、496例LUAD完整临床病理资料,分析LYPD3的表达与患者临床特征的关系,采用COX回归模型进行多因素生存预后分析,通过基因集富集分析相关信号通路。结果LYPD3在LUAD组织中高表达,与人种、肿瘤分期及生存状态相关(P=0.0448、0.0191);LYPD3表达及cg25340403甲基化是影响LUAD预后的独立因素;LYPD3通过调节P53信号通路影响LUAD患者的发病及预后。结论LYPD3在LUAD中表达上调,其过表达与不良预后相关,且LYPD3可能通过P53途径促进LUAD的进展。Objective To investigated the role of LYPD3 in the patients with lung adenocarcinoma(LUAD)and its value for predicting clinical prognosis.Methods The related data were obtained by retrieving the Cancer Genomic Atlas(TCGA):the LYPD3 gene expression and DNA methylation expression situation in 535 cases of LUAD tissue samples and 59 cases of paracancerous tissues sample,complete clinicopathological data in 496 cases of LUAD.Then the relationship between LYPD3 expression and clinical features of the patients was analyzed.The COX regression model was adopted to analyze the multiple factors survival prognosis.The related signal pathway was analyzed by the gene set enrichment analysis.Results LYPD3 was highly expressed in LUAD tissue and was correlated with the race,tumor stage and survival status(P=0.0448,0.0191).Furthermore,the LYPD3 expression and cg25340403 methylation were the independent factors affecting LUAD prognosis;LYPD3 affected the onset and prognosis of LUAD patients.Conclusion The LYPD3 expression is upregulated in LUAD,its overexpression is correlated with adverse prognosis,moreover LYPD3 may promote the LUAD progress by p53 pathway.
